Objective: This study through the preparation of type1diabetes rats model which ratscaused by high blood sugar oxidative stress state, observe the change of electricalremodeling in rat myocardial cells under oxidative stress state, and applies the proprietaryChinese medicine "shensong yangxin capsule" to intervene, to explore the mechanism ofoxidative stress state electrical remodeling in myocardial, and watch the proprietary Chinesemedicine "shensong yangxin capsule" the effect of intervention myocardial electricalremodeling in rats, to provide experimental basis for the clinical application of TCMprevention and treatment of arrhythmia.Methods:1. Choose the weight is220+30g SD male rats (clean), all the rats byadaptive feed l weeks, were randomly divided into two groups: normal control group (n=10)and artificial for diabetic model group (n=20).2. The streptozotocin (STZ) soluble in sterilization tendency for0.1mmol/L pH=4.2citric acid buffer, the concentration of1%, according to45mg/kg intraperitoneal injection ofone-time;Normal control group rats by intraperitoneal injection of one-time volume such ascitric acid buffer.One week after determination of caudal vein blood sugar, random bloodglucose is greater than or equal to16.7tendency/l as diabetic rats in experimental rats.3. The DM were randomly divided into two groups, diabetic group (n=9) and andshensong yangxin intervention group (n=9), among them and shensong yangxin interventiongroup according to0.6g/kg plus3ml distilled water, mixture liquid filling,1times a day,12consecutive weeks; The rest of the group were given the same volume of physiologicalsaline lavage.In diabetic rats model building successful randomized4weeks,8weeks,12weeks after determine the weight and blood glucose, serum SOD, MDA concentration, checkthe echocardiogram, determination of the left ventricular end-diastolic diameter (LVDd) andejection fraction (LVEF), and the ventricular electrical stimulation in week12recordsventricular muscle effective refractory period (ERP) and monophasic action potentialduration(MAPD) After that, we take the myocardial tissue pathology examination. Results:1. Preparation of STZ diabetic rats simulated test success rate of90%.2. Thegeneral situation is: oxidative stress and and shensong yangxin group rats weight is lowerthan the normal control group, the blood sugar is higher than the control group, thedifference was statistically significant (P <0.01);Oxidative stress group and the rat bodyweight, blood sugar and shensong yangxin group has no statistically significant difference(P>0.05).3. The oxidative stress indicators: and shensong yangxin group blood SOD theoxidative stress concentration is high, but slightly lower than normal control group, thedifference was statistically significant (P <0.01);And shensong yangxin group was lowerthan those of oxidative stress group, the MDA concentration higher than normal controlgroup, the difference was statistically significant (P <0.01).4. In the group of shensongyangxin ERP and MAPD shorter than oxidative stress group (P <0.01), compared withnormal control group there was no significant difference, no statistical significance.5.Compare cardiac function, LVEF in shensong yangxin group is slightly high oxidative stressgroup, but was lower than those of normal control group, the statistical difference wasstatistically significant (P <0.01); Compared with the oxidative stress group, the LVDd inshensong yangxin group relatively shorter, but slightly longer than normal control group,statistics difference was statistically significant (P <0.01).6. Pathology results: comparedwith control group, the oxidative stress group rats myocardial cell edema, the nucleusincreases, horizontal stripes disappear, broadening gap, compensatory hypertrophymyocardial cells, muscle fibers decrease, disordered arrangement of myocardial fibers andmuscle bundle edge is coarse, dissolve more visible in muscle and muscular rupture;Massonstaining showed cardiac collagen tissue levels increased significantly, transverse section ofthick collagen fiber are connected into a net, heart came acute interstitial fibrosis.Conclusion: High blood sugar induced oxidative stress state can promote or myocardialcell electrical remodeling and structural reconstruction;And shensong yangxin capsule hasgood resistance to oxidative stress effect, can improve the activity of serum S0D, to reducethe concentration of serum MDA, inhibiting myocardial cell electrical remodeling andstructure remodeling, and can improve heart function.
