ObjectiveOur country was the country which there were the most abundant resources of wild mushrooms. Wild mushroom resources in Northeast China Region were very rich, especially in the southeastern Jilin Province, which there was a vast complex high coverage the Changbai Mountain area. There were very good conditions for the growth of wild edible mushrooms which were offered by its climate. The area of edible and medicinal mushrooms in Changbai Mountain was up to200kinds. However, due to mushroom preservation and processing technology is not mature enough so the development of the mushroom is still relatively low utilization, the development is still in its infancy, low processing product technology content, high added value. In this study, polysaccharide extracted from five wild edible mushroom in Changbai Mountain area was choosen to measure the antioxidant and anti-mutant, then therr would be a theoretical basis for the development of edible mushrooms in health products and precision machining.Methods1.After water extraction and alcohol precipitation mushroom extract polysaccharides and mushroom polysaccharide content was measured by phenol sulfuric acid method of polysaccharide. By chromogenic assay Congo red mushroom polysaccharide content in the main activity of the polysaccharide dextran.2.To study buckwheat mushroom polysaccharide antioxidant activity, experiment with different concentrations of the five mushroom polysaccharides as the research object, were carried on mushroom polysaccharides on DPPH radical scavenging and hydroxyl radical scavenging ability.3.Acute toxicity by oral LD50tests were measured values for toxicity classification. Ames test method using a modified,micronucleus test in mouse bone marrow cells were observed mutagenicity and anti-mutagenicity mushroom polysaccharides. Anti-mutagenic effects of mushroom polysaccharides study provides a theoretical basis for the continued development of the health effects of wild edible fungi.Results 1.Phellinus igniarius, glossy ganoderma and Inonotus higher polysaccharide content of over60%; higher glucan content Inonotus of over30%. Which accounted for69%of Hericium Hericium glucan polysaccharides, Ganoderma lucidum and Phellinus and accounted for less glucan polysaccharide content.2.Five kinds of mushroom polysaccharides There was some eflect on DPPH radical scavenging, but there were some differences in the ability to clear the size of Polysaccharids on DPPH radical scavenging is better than the other polysaccharides. Polysaccharide concentration in125μg/mL clearance rate wsa up to83.8%.Mushroom polysaccharides on hydroxyl radical scavenging rate, was up with the increasing concentration, but the trend was not obvious. In other mushroom polysaccharide concentration of hydroxyl radical500μg/mL clearance rates were between30.1%-42.9%, showing some of the hydroxyl radical scavenging ability.3.Anti-mutation test:five kinds of edible mushrooms in MNNG (0.4μg/plate) induced TA100test, anti-mutation at the highest concentration of200μg/plate of conditioning have reached over70%. TA98experiment in4NQO induced the highest concentration of200μg/plate Inonotus showed more than80%inhibition, and in TA100experiment Phellinus and Inonotus inhibition rate reached90%. Intake of five kinds of mushroom polysaccharides have no effect on micronucleus formation, indicating that the five mushroom polysaccharides not mutagenic. Which Inonotus polysaccharide Ganoderma lucidum polysaccharides in the500mg/kg BW concentrations of mouse bone marrow micronucleus effect best, the inhibition rate reached77.0%and70.1%.Conclusionl.Five kinds of mushroom polysaccharides ther were some antioxidant, which Polysaccharide antioxidant capacity than other mushroom polysaccharides.2.Five kinds of mushroom polysaccharides have certain anti-mutagenic, which Polysaccharide and Polysaccharide Inonotus antimutagenic effect was strongest.3.Five kinds of mushroom polysaccharides on mouse micronucleus formation inhibition of the mutant has a preventive effect, which Inonotus polysaccharide shows the strongest inhibitory effect.
