ObjectiveIn this study, Shantou, Guangdong Province, namely four years of age, five, eight, twelve, fifteen years of incense herbs samples for chemical and analgesic efficacy studies, A comparative study of the above two aspects to investigate the influence of age on the quality of incense herbs, thus incense herbs evaluate the quality of different ages, to further establish the quality evaluation system and industrial research systems to provide ideas for incense herbs.Methods1. Using the2010version of "Chinese Pharmacopoeia" in the entry under aloes character identification, TLC, alcohol-soluble extract of the content and micro-sublimation experiment, as well as UV-identification method to identify its quality.2. According to general quality of research methods2010version of "Chinese Pharmacopoeia"(a) under Appendix.3. Using a variety of reagents for identification of incense herbs extract parts of each ingredient systematic research.4. Orthogonal extraction process incense volatile components, and volatile components by GC-MS analysis.5. Using a hot plate, acetic acid-induced writhing test in mice pain analgesic efficacy study situation is different herbs incense age of volatile components, and to explore the relationship between the analgesic effect of herbs and incense between the volatile components. Results1. Characterization results show that:four ages of Aquilaria agallocha characters, TLC and trace sublimation experiment results accord with Pharmacopoeia;with the increase of the content of old growth Aquilaria alcohol extract of herbs, color deepened, UV absorption increases.2. The results show that the general quality:four different samples of ash and old incense herbs extract content increased with increasing age growth, and are quite different, especially alcohol-soluble extract (hot dip method) and ether-soluble extractives (cold soak France) content varies greatly.3. Chemical composition of the system pre-experimental results show:the experimental samples containing organic acids, volatile oils, phenols, tannins, sugar and sugar, polysaccharides and glycosides, steroid saponins, sterols, coumarin and glycosides, lactones, strong cardiac glycosides; may contain alkaloids, amino acids, peptides, triterpenoid saponins, proteins; contain flavonoids and anthraquinones.4. Ultrasonic Extraction Process Experimental results show that:The best extraction process:30ml chloroform at room temperature soak6h, ultrasonic extraction20min.GC/MS analysis showed that:44samples were detected chemical composition,5years,8years,12years,15-year-old incense herbs isolated89,88,62,65each sample peaks were detected28,20,29,31compounds. Isolated from each sample chemical composition is slightly different, but all sesquiterpenes, mainly composed of aromatic and contain aloes characteristic ingredient benzyl acetone,β-guaiac ene, grapefruit ketones. With the growth of compounds in age sesquiterpene there was increased after the first reduction in the downward trend, the opposite was the first increase in aromatic compounds and then decreasing trend.5. Mouse hot plate induced pain Experimental results show that:the experimental group could extend mouse hot plate pain threshold, analgesic efficacy is superior to the control group.8years,12years, students from medicine group effect faster than the five-year,15-year-old medicine group and at90min after efficacy was significantly better than the5-year,15-year-old medicine group (P<0.05, P<0.01),8-year-old medicine group pain threshold at each time point were more than12years of raw medicinal herbs group, but the difference was not significant (P>0.05), four eight-year-old born in medicinal herbs group best analgesic efficacy. The5-year and15-year-old pain medicine group cross the threshold changes, analgesic efficacy difference was not significant (P>0.05). Mouse acetic acid writhing test results showed that:after the administration could significantly reduce the30min each experimental group of mice writhing (P<0.01), analgesic efficacy is superior to the control group.8years,12-year-old writhing medicine group was significantly less than5years,15-year-old medicine group (P<0.01), raw medicinal herbs group eight years writhing than raw herbs group for12years, four old medicine in8-year-old medicine group the analgesic efficacy best.15-year-old medicine group writhing less than5years old medicine group, but not significantly (P>0.05). Conclusion1. Four ages of Aquilaria agallocha are authentic Aquilaria agallocha;Aquilaria agallocha ethanol extracts content, color, UV absorption values were closely related to age.2. Four ages of Aquilaria agallocha’general quality was positively related with age, and in every age differences.3. Although the chemical composition of the system can not pre-test the chemical composition of herbs contained an accurate characterization, but with guidance for post-study.4. Analysis on the change trend of all kinds of components by GC/MS, relativeamount of each are not identical, and lack of positive correlation with tree age. Prompt detection can increase the above related projects in Aloes quality standards, in order to better control the quality.5. Aloes analgesic efficacy is not positively correlated with the age.8years,12-year-old medicine analgesic efficacy than5years,15year old,8years old the best analgesic efficacy of incense herbs. Contact GC/MS analysis,8years aloes contents contains aromatic compounds were the most, basic substances that aloes in aromatic compounds may contain its analgesic effect, the specific matter needs further study.
