Objective:The research used CCl4-induced acute liver injury model in mice, to investigate the protective effects of Taiyinmahuang decoction on the acute liver injury and to explore its possible mechanism.Methods:The sixty Kunming mice were divided randomly into six groups:the normal control group, theCCl4model group, the positive control group, Taiyinmahuang decoction high dose; medium dose, and low dose groups. Before the trial, all the mice were fed adaptively3days and then were intragastric administrated. The mice in normal control and CCl4model group were daily intragastric administrated with the same volume of sterilizing saline,the mice in the positive control groupwere intragastric administrated with Bifendate150mg/kg, the dosage of Taiyinmahuang decoction high dose, medium dose, and low dose group was2.00g/kg,1.00g/kg and0.50g/kg respectively, all the medications were given at a volume of0.2ml/10g, Once daily for seven consecutive days. on the7th day,1h after the intragastric administrated,addition to the mice of the normal control group were set up by giving the same volumn intraperitoneal injection of saline, the mice of other groups were set up by giving intraperitoneal injection of0.2%CCl4peanut oil liquid (10ml/kg),the mice should be killed after24h of forbidding to provid food but water. After the preparation of samples, detecte the activity of mouse serum alanine aminotransferase (ALT) and aspartate aminotransferase(AST),the content of liver homogenates malondialdehyde (MDA)、superoxide dismutase(T-SOD) and the activity of glutathione peroxidase(GSH-PX).Results:Compared with the model group, Taiyinmahuangdecoction can significantly suppress levels of rised ALT, AST which caused by CCl4of liver damage, reduce the level of rised MDA in liver homogenates, enhance activities of T-SOD and GSH-Px, that high dose group best effect.Conclusion:Taiyinmahuangdecoction has significant protective effect on CCl4-induced acute liver injury in mice,its mechanisms might be associated with scavenging oxygen free radical、inhibiting the lipid peroxidation and enhancing the antioxidant capacity.
