| Research purposes:The purpose of the issue is using carbon tetrachloride (CC14) to produce the model of acute liver injury, and then study the protective effect of Korean medicine "bear bile powder" for acute liver injury, and to explore the role of the bear bile Powder mechanisms.Research methods:Dividing 60 healthy male Kunming mice into 6 groups randomly, which is positive contradistinguish group and normal contradistinguish group, model of CC14 group and the bear bile high dose, medium dose and low dose group. Before starting the experiment, the mice will be feeded adaptivly for three days, and then start intragastrical administration. CC14 model group and the normal control group fed daily with an equal volume of Sterile saline, the positive control group was given Ganlixin 0.2ml/10g gavage, the dosage of bear bile powder high dose, medium dose and low dose group were:45mg/ml,15mg/ml,5mg/ml, dosing volume is 0.2ml/10g, feeding once daily, and keep the operating for seven days. After gavage one hour in the seventh day, the normal control mice received an equal volume of saline, and the remaining five groups are injected 0.2% carbon tetrachloride in intraperitoneal with 0.2ml/20g, fresh off and free access to water for 24 hours, sacrificed model mice by cervical dislocation. Then detect serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), liver tissue glutathione peroxidase (GSH-Px) and malondialdehyde (MDA).the total of superoxide dismutase (T-SOD).Research results:Comparing with the model group, each dose group of bear bile powder can significantly reduce elevated glutathione peroxidase (GSH-Px) and two kinds of aminotransferase in the serum of experimental liver injury mice caused by CC14, and decrease liver homogenates increased malondialdehyde (MDA), increasing the content of superoxide dismutase (T-SOD).Research conclusion:Korean medicine "bear bile powder" can protecte acute liver injury in mice which caused by the CC14, the mechanism may be enhance antioxidant ability, inhibit lipid peroxidation and remove oxygen free radicals. |
