Objective Exploring the protective effect of curcumin on acute liver injury used by combination of alcohol and carbon tetrachloride.Method Prepare mice acute liver injury model. Fifty healthy male BALB/C mice were randly divided into5groups (n=10in each group). Control group: Intraperitoneal injection of olive oil,tail intravenous injection of PBS, intragastric administration with glucose. CCl4treated group:Intraperitoneal injection of CCl4, tail intravenous injection of PBS, intragastric administration with glucose. CCl4plus alcohol treated group:Intraperitoneal injection of CCl4, tail intravenous injection of PBS, intragastric administration with alcohol. Low doses of curcumin treated group: Intraperitoneal injection of CCl4, tail intravenous injection of1×10-4M curcumin, intragastric administration with alcohol. High doses of curcumin treated group: Intraperitoneal injection of CCl4, tail intravenous injection of3×10-4M curcumin, intragastric administration with alcohol. All animals were killed at the end of two weeks’s treatment. Detect the biochemical criterion of ALT, AST, Alb and TBIL in the animal model; Observe the pathology change of hepatic tissue by HE staining and Masson staining; Detect the expression level of tumor necrosis factor TNF-a, TGF-P and a-SMA by Western Blot technique; Observe expression of CHOP by immunofluorescence staining; Observe ultrastructure of hepatic cells by transmission electron microscopy.Results Compared with control, in CCl4model group the serum levels of AST,ALT,TBIL were significantly elevated (P<0.05), Alb was significantly reduced (P<0.05);HE staining and Masson staining showed the liver injury; Western Blot showed enhanced expression of TNF-a, TGF-β and a-SMA. In CCl4plus alcohol treated group the serum levels of AST,ALT,TBIL were significantly elevated(P<0.05),Alb was significantly reduced (P<0.05); HE staining and Masson staining showed serious liver injury, interlobular septal apeared; Western Blot showed higher expression of TNF-a, TGF-β and a-SMA. In high doses of curcumin treated group AST, ALT and TBIL were significantly reduced and the formation of collagen fibers was inhibited; The expression of TNF-a, TGF-β and a-SMA was significantly reduced. Compared with CCl4plus alcohol treated group it was not obvious in low doses of curcumin treated group.Immunofluorescence showed high expression of CHOP in CCl4plus alcohol treated group, high doses of curcumin significantly reduced the expression of CHOP. Electron microscopy showed the endoplasmic reticulum is badly damaged in CC14plus alcohol treated group and high doses of curcum can improve it.Conclusion Curcumin can reduce the activation of HSC and the production of EMC by reducing the expression of TNF-α, TGF-β and α-SMA, thus reduce or prevent the development of liver fibrosis. Curcumin can provide protection for CCl4and alcohol-induced liver injury by reducing the endoplasmic reticulum stress.
