ObjectivesThis study chose rs266729, rs7649121two SNP sites as the main research target, from the plasma level and gene level to discuss the relationship between adiponectin and hypertension, diabetes.MethodsA case-case study was designed in this study. A total of494Han Chinese subjects were recruited into this study, including130control subjects with neither diabetes mellitus nor hypertension,107cases who had DM alone,143cases who had hypertension alone,114cases with both DM and hypertension.All objects liver and kidney function is normal, and are from the han people in tianjin region. Automated Bilchemical Analyzer was used to measure the concentration of serum CHO、TG、 LDL—C、HDL—C、FPG、2hPG. The serum adiponectin level was measured by ELISA. Genomic DNA were extracted from the blood cell. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RPLP) were used to detect the adiponectin gene SNP rs266729、rs7649121Results1、EH group of patients age, BMI, TC, systolic pressure, diastolic blood pressure levels are significantly higher than control group (P<0.05), And SBP, DBP level is significantly higher than that of DM group; the DM patients with BMI, TC, TG, LDL-C,FPG,2hPG, HbAlc levels are significantly higher than control group (P <0.05), and FPG,2hPG,HbAlc levels are higher than EH group; EH and DM group of patients with BMI, SBP, DBP, TG, LDL-C,FPG,2hPG, HbA1c levels are higher than the control group (P<0.05), plasma HDL levels lower than the control group (P<0.05), at the same time BMI, TC, FPG,2hPG, HbAlc is higher than EH group, BMI, SBP, TC levels higher than that of DM group, the difference was statistically significant.2、other group compared with control group, serum adiponectin levels are lower than the control group, the difference is statistically significant (P<0.05), hypertension, diabetes mellitus group serum adiponectin level is lower than simple diabetes group and simple hypertension group, the difference statistically (P<0.05).Hypertension and type2diabetes group serum adiponectin level had no significant difference(P>0.05).3、BMI,TC is a risk factors for hypertension, BMI, LDL-C is a risk factor for diabetes, APN for hypertension diabetes protection factors.4、rs266729allele frequency and genotype distribution between hypertension group and non hypertension group with significant difference, And the Logistic regression analysis showed that rs266729locus of CG genotype and G allele carriers (CG+GG) are the protective factors of hypertension. The rs7649121allele and genotype distribution frequencies in non hypertension group and hypertension group and non diabetes group and diabetes group had no obvious difference, and Logistic regression analysis showed that the rs7649121polymorphism with hypertension and diabetes mellitus have no obvious correlation. Adiponectin SNPrs266729, rs7649121loci were not significantly associated with adiponectin plasma levels.Conclusion1、low adiponectin levels may be a risk factor for hypertension and diabetes.2、Adiponectin gene rs266729polymorphism may be associated with hypertension in Tianjin area, which can reduce the incidence of hypertension. And Adiponectin gene SNPrs7649121polymorphism with hypertension and diabetes independent.
