Objective Investigate the relations among the transcription factor7-like2(transcription factor7-like2, TCF7L2) gene Single Nucleotide Polymorphisms (SNP), essential hypertension (EH) and the type2diabetes mellitus(T2DM) in Yanbian Chinese Korean and Han populations. Methods Use of single-base extension assay SnaPshot to detect TCF7L2gene SNP (rs11196205, rs11196218) genotypes.Total181case T2DM were collected with93Chinese Korean and88Han,99EH case also collected with49Chinese Korean and50in Han, Normal Glucose Tolerance (NGT)group collect83Chinese and99Han with182case in total.Results1.The frequencies of the allele and the genotype of the rsl1196205, rs11196218TCF7L2are in line with Hardy-Weinberg equilibrium (p>0.05), in T2DM group, EH group and NGTT group; The rs11196205GC genotype frequencies of the Han (0.172) is significantly higher than Chinese Korean (0.072),The frequencies of the allele and the genotype of rs11196218didn’t show significant differences between the Chinese Korean and Han (p>0.05).2. In the Han group, rs11196205-C GC genotype frequencies of T2DM patients is significantly lower than NGT group(0.057VS.0.080,p=0.031);There no any difference between the allele frequencies of rs11196218polymorphism and genotype frequencies (p>0.05) in three group of both Chinese Korean and Han.3. Haplotype CA showed negative correlations with susceptibility of EH in Yanbian-Korean (p=0.037, OR<1).4. The WHR levels of the phore individual with genotype GC or CC are significantly lower than the phore individuals with genotype GG in three group (p<0.05) of Han nation, BMI levels of the phore individual with genotype GC is significantly lower than the phore individual GG in EH group(p<0.05), INS levels of the phore individual with GC/CC is significantly higher than individual with GG (p<0.05) in T2DM group, it show the characteristics of dominant inheritance mode;The correlation relation between various genotypes rs11196218polymorphism and clinical indexes (p>0.05) not found.5.The allele C frequency of rs11196205in Yanbian-Korean and Han was Significantly lower than CEU/YRI (p<0.01); The allele A frequency significantly of rs11196218in Korean was Significantly higher than YRI (p<0,01).higher than JPT (p<0.05); in Han was Significantly higher than YRI (p<0.01),higher than JPT (p<0.05), but lower than HCB (p<0.05).Conclusion1.TCF7L2gene rs11196205-C may be protection factor of T2DM and EH group in Yanbian-Han, it may be effect the INS level lead to control diet and reduce the risk of T2DM and EH;2.The polymorphism of rs11196205may not the susceptible gene of T2DM and EH in Yanbian-Korean;3.The polymorphism of rs11196218does not have a relation with the T2DM and EH of the Yanbian-Korean and Han;4.Haplotype CA is a risk haplotype of EH in Yanbian-Korean population;5.The polymorphism of rs11196205has difference in different nationality and race. |
