Objective: This study is to record the gene test results of the tumor, which is based on lung cancer xenotransplanted animal models with individual characteristic, hoping to understand whether the gene mutations in two lung cancer remain consistent.Methods:Collected25cases lung cancer patients underwent surgery in Tianjin Chest Hospital in March2012-in june2012,to get fresh lung tissue specimens in six hours and use vaccination needle planted in their BALB/C nude mice subcutaneous tissue,tumorigenicit by the first generation,take the first generation of tumor tissue transplantation model,repeat the above steps until a fifth generation of animal models.Then extract DNA from the10cases of successful modeling tumor xenograft tissue and tumor source according to the instructions of DNA extraction kit, then go to genetic testing about the source of tumor xenografts by PCR and gene sequencing technology.Results:Accordig to gene test result of10source and xenografts the mutations in different ung cancer patients n the same istological are different,No.156,No.369,No.382,No.781transplanted tumors are squamous cell carcinoma,which of No.156is MET and TP53mutant,369and781were both TP53mutant,No.382is the wild type.No.368and No.959are mixed adenocarcinoma,which of No.368is EGFR and TP53mutant and No.959is KRAS and TP53mutant; gene mutation about tumor and xenograft tumor in lung cancer cases can basically remain consistent,inconsiitent mutation after only two cases,in which the No.959tumor source is KRAS wide-type and xenograft is mutation-type,No.369TP53tumor tissue as a source of wide-type, xenograft is mutant-type.Conelusion:The method of xenotransplanting individual characterized human lung cancer tumor to immune deficient mice is easy,reliable and repeatable. and the gene mutation is different as well.It proves that the tumor model we have established have individual characteristics.The xenografts can save the most important tumor gene in primary lung cancer and only a small number of genes mutated in passaging process,its fidelity is better to prove that lung cancer xenografts can be used as tools for individualized trement.
