Backgroud and purpose:mTOR was an important gene for regulating the cell growth and cell proliferation. mTOR could activated downstream related genes.PTEN was a negative regulated gene in signaling pathway, it could restrained the activation of mTOR. To detect the expression of mTOR and PTEN in non-small cell lung cancer(NSCLC), and explore their role on prognosis of patients with NSCLC.Mehtods:The samples of cancer tissues and normal lung tissues from 78 patients with NSCLC, were detected the expression of mTOR and PTEN by the Real Time-PCR and immunohistochemistry. The difference of mTOR and PTEN expression was compared by Student's t-test. Cox regression model was used to analyze the relationship between the influencing factors and the progrosis. Kaplan-Meier survival curves and the log-rank test were used to analyze progression free survival.Results:The levels of mTOR expression in NSCLC tissue was significantly higher than that in normal lung tissue, while the levels of PTEN expression in NSCLC tissue was significantly lower than that in normal lung tissues(P<0.05). There was no significant correlation between mTOR and PTEN expression and patients'age, gender, pathological type, differentiation, lymph node metastasis, distant metastasis, except T stage. mTOR and PTEN expression had important impact for the progrosis.Conclusion:The absent and/or low expression of PTEN and the activated mTOR may play an important role in development of NSCLC. Absent and/or low expression of PTEN and high expression of mTOR may be promising for poor prognosis of patients with NSCLC. Backgroud and purpose:Pax-6 gene was a transcriptional factors,which participated human embryo development .It can regulate and control Proliferation of embryo cells.According to previous research report, Pax-6 was high expression in cerebral cancer and breast cancer.But, it was low expression in Prostate Cancer. Pax-6 had some diverse characteristics and functions in diverse tumors.until now, the expression features and functions of Pax-6 was unknown This experiment mainly discusses the expression of Pax-6 genes in NSCLC and proliferative ability in NSCLC cell lines.Mehtods:The samples of cancer tissues and normal lung tissues from 55 patients with NSCLC, were detected the expression of Pax-6 by the Real-Time PCR . 8 lung cancer cell lines and a normal cell line were detected the expression of Pax-6 by the Real-Time PCR . After silence Pax-6 genes by RNAi technology, we researched the proliferation ability and migrating ability of H1299,A549 cellsResults:Pax-6 gene mRNA was high expressed in NSCLC tissue and lung cancer cell lines.. After silence Pax-6 genes by RNAi technology, the proliferation ability was significant restrained, and but it had no significant effect on the migration ability of H1299 and A549 cells.Conclusion: Pax-6 gene mRNA was hing expressed in NSCLC tissue and lung cancer cell lines. Pax - 6 paly a improtant role in NSCLC cell proliferation.
