[Objective] To study the composition of propolis antibacterial effective part from Chuxiong Yaoan,Yunnan Province,extraction and isolation of it. The effects of different components of the Propolis antibacterial effective parts on4main periodontal pathogens in vitro were investigated. To find preliminary effective antibacterial component of propolis.[Methods]1. Using high-performance liquid chromatography to analysis propolis antibacterial effective part and the standard reference of Caffeic acid, Toadflax flavin, Pinocembrin, Alpinetin. According to the results of HPLC, using ODS column chromatography to extract and isolate the propolis antibacterial effective part.Then use the Thin layer chromatography to combine and concentrate the parts which hane the same Rf,and divided propolis antibacterial effective part into A, B two components.2.By anaerobic culture main periodontal pathogens, Pg, Pi, Fn and Aa standard strains.The standard strains were tested by the slip diffusion method. Thetesting groups were5%,2.5%EEP, Caffeic acid, Pinocembrin, componentsA,B of propolis antibacterial effective part.Metronidazole(MNZ) solution of0.05%was used as positive testing group,95%Methanol and ethanol was used as negative tesing group.[Results]1.HPLC’s chromatogram of propolis antibacterial effective part and standard reference indicated that propolis antibacterial effective part contains Caffeic acid and Pinocembrin,excluding Toadflax flavin and Alpinetin.According to the result, using ODS column chromatography and Thin layer chromatography divided propolis antibacterial effective part into components A that contains Caffeic acid and components B that without Caffeic acid.2.The antibacterial effect of four kinds of bacteria, components A、B and Caffeic acid on4periodontal pathogens in certain concentrations within the scope of inhibition, and its no inhibition of Pinocembrin. in the same concentration, the antimicrobial activity of propolis antibacterial effective part was stronger than each monomer components and mixtures.The difference was statistically significant (P<0.05).[Conclusion]1. Using high-performance liquid chromatography to analysis the composition of propolis antibacterial effective part from Chuxiong Yaoan,Yunnan Province and it is indicated that propolis antibacterial effective part contains Caffeic acid and Pinocembrin,excluding Toadflax flavin and Alpinetin. By ODS column chromatography, TLC combining HPLC methods to achieve the purpose that initial separation of propolis antibacterial effective part.2. in the same concentration, the antimicrobial activity of propolis antibacterial effective part was stronger than each monomer components and mixtures. The antibacterial effect of propolis may be the result of multiple component synergy.
