[Aim] To study the antimicrobial effect of propolis packed with chitosan gel on Porphyromonas Gingivalis (Pg), Actinobacillus Actinomycetem (Aa)、Prevotella intermedia (Pi) and Fusobacterium nucleatum (Fn) in the periodontitis model built on Beagle dogs.[Methods]1. Periodontitis modeling:3adult Beagle Dogs (male) raised in3separated coops, on which ligature-induced periodontitis will be built, check periodontal indexes and X-ray on a time basis.2. Primer design for Pg, Aa、Pi、Fn and IC, PCR tests the amplification efficacy and specificity of primers, to detect4target bacterias.3. Real-time PCR standard curve of4target bacterias and IC made by Gel extraction, correlations of all standard curve are beyond0.98, efficacy is within90%-120%, which is compliance with criteria of qualified primer. Real-time PCR detects the number difference of4target bacteria pre-and-post-drug delivery, test the effect of propolis on4target bacteria, and compare the change of PD and AL after drug delivery.[Results]1. Related periodontal indexes and X-ray indicate periodontitis model has been built up.2.Distinguishable bands are shown in gel electrophoresis, the size of the bands match the size expected, no apparent primer dimers are found.3. Good correlations are seen in the standard curve (correlations are beyond0.98), efficacy of Real-time PCR is within90%-120%, which is compliance with criteria of standard curve. Real-time PCR detect shows no significant difference among Pg and Pi groups, P>0.05; significant difference is found between propolis group and negative control group (P=0.011) of Aa in1st week and3rd week drug delivery. Statistical difference is found between propolis group and negative control group (P=0.025) of Fn in24hours drug delivery. Statistical significance is shown in PD and AL change after drug delivery finished.[Conclusion]1. Periodontitis model in beagle dogs is made successfully by ligature around the cervical of chosen teeth, beagle dog is suitable for research related to periodontitis.2. Specificity and efficacy of primers are qualified the primer expectation, expected bands are got,4target bacterias existing are detected by PCR.3. Propolis packed with chitosan gel shows no suppression effect on Pg and Pi, it shows some prohibition effect on Aa and Fn. Propolis is effective in treating experimental periodontitis according to the changes in PD and AL after drug delivery.
