【Objective】This thesis focused on the extraction and isolation of the raw Propolis from Yunnan Province,and the contents of flavonoids and total polyphenols in different components were determined by ultraviolet spectrophotometer. The effects of different components of the Propolis on4main suspected periodontal pathogens in vitro were investigated.To find preliminary effective antibacterial component of propolis and preliminary analysis of the components.【Methods】1.Crude extract Propolis were obtained95%ethanol as solvent by Ultra-sound method. By polyamide column chromatography for separation and purifyca-tion of crude extract of propolis to be different parts of Propolis (FJ-00, FJ-O1and FJ-02).Using rutin and Protocatechuic acid respectively as standard samples for the determination of flavonoids and total polyphenols in different components were determined by ultraviolet spectrophotometer.2. By anaerobic culture main suspected periodontal pathogens, Pg, Pi, Fn and Aa standard strains.The standard strains were tested by the slip diffusion method. Thetesting groups were EEP which concentrations were20%.10%,5%.2.5%.1.25%,0.625%. Metronidazole(MNZ) solution of0.05%was used as positive testing group and95%ethanol was used as negative tesing group.3. Using high-performance liquid chromatography with electrospray ion trap mass spectrometry for effective antibacterial component of chemical components in propolis for preliminary analysis.[Results] Different components of the Propolis were obtained by Ultrasound method and polyamide columnand chromatography for separation and purification.Flavonoid content of the each component which was measured by UV spectrophotometry was FJ-002.25%, FJ-013.45%, FJ-021.5%.Total polyphenols content of the each component was FJ-003.6%,FJ-014.8%,FJ-022.8%.2. The antibacterial effect of four kinds of bacteria, in the same concentration, antimicrobial activity of FJ-01parts was significant difference statistically comparing with other parts (P<0.05). on the whole, the same concentration, and antimicrobial activity of FJ-01was stronger than the other two parts.3. By high performance liquid chromatography with electrospray ion trap mass spectrometry on the major effective antibacterial component did a preliminary study on the chemical constituents of FJ-01, came mainly in the FJ-01part may have caffeic acid, caffeic acid ester compounds, flavonoids, two hydrogen hydrogen II yellow ketone alcohol and (or) the second hydrogen chalcone class, especially of Jack Pine of derivatives.【Conclusion】1. Different components of the Propolis from Yunnan Province were obtained by Ultrasound method and polyamide columnand chromatography for separation and purification. Flavonoid content of the each component which was measured by UV spectrophotometry.The methods are simple, low-cost and accurate. Flavonoid content of the each component which was measured by UVspectro-photometry was FJ-002.25%, FJ-013.45%, FJ-021.5%. Total polypenols content of the each component was FJ-003.6%,FJ-014.8%,FJ-022.8%.2. Obtained by using polyamide column chromatography separation of yaoan three parts of propolis on Pg,Pi, Fn, Aa in certain concentrations within the scope of inhibition,inhibitory effect of four kinds of bacteria, showing obvious relationship between concentration. when at the same concentration. bacteriostatic effect stronger than FJ-00. FJ-02, FJ-01.3. On the analysis of the effective fraction of propolis results FJ-01part may have caffeic acid, caffeic acid ester compounds, flavonoids, two hydrogen hydrogen II yellow ketone alcohol and (or) the second hydrogen chalcone class. especially of Jack Pine of derivatives.
