Objective: To study the expression and significance of p65, VEGF and MMP-9protein in osteosarcoma and its correlation with clinicopathologic features inosteosarcoma.Methods: The expression of P65, VEGF and MMP-9proteins were determined by theimmunohistochemical staining. The relationship between these three proteins andclinicopathologic features in osteosarcoma was analyzed by χ2test and Spearman’s rankcorrelation analysis. Kaplan-Meier‘S method was used to analyze the survival.Results:(1) In45cases of osteosarcoma, the positive rates of p65, VEGF and MMP-9protein were62.2%,75.6%,46.7%, respectively,(p<0.05).(2) The positive rate of p65correlated with poor tumor differentiation and late Enneking stage,(3)VEGF protein expression was significantly higher in larger tumor size (morethan5cm)(P <0.05). In Enneking staging, VEGF protein expression level increased withthe progress of the stage (P <0.05). The expression of VEGF protein in the tumor ofstage III, Ⅱ B is significantly higher than in the tumor of type IIA (P <0.05).(4) The expression of MMP-9protein in poorly differentiated tumors is higher thanthe high differentiation of tumor (P <0.05). In Enneking staging, MMP-9proteinexpression level increased with the progress of the stage. The expression of MMP–9inthe tumor of stage III, Ⅱ B is significantly higher than in the tumor of type IIA (P <0.05). The positive rate of MMP-9correlated with lung metastasis(p<0.05).(5) In45cases of osteosarcoma, the positive cases of p65expression shew VEGF in25cases. The negative cases of p65expression shew negative VEGF expression in8cases.The above groups had positive correlation between (P=0.005, r=0.410). In45cases ofosteosarcoma, the positive cases of p65expression shew MMP-9in17cases. The negativecases of p65expression shew negative VEGF expression in13cases. The above groupshad negative correlation between (P=0.005, r=0.005). (6) The average survival time of and the5-year survival rate the positive group of P65were32months (95%CI:23.1,40.9) and25.0%. The average survival time of and the5-year survival rate the negative group of P65were56months (95%CI:54.3,57.7) and76.5%. The average survival time of and the5-year survival rate the positive group ofVEGF were32months (95%CI:23.8,40.2) and35.3%. The average survival time of andthe5-year survival rate the negative group of VEGF were51months (95%CI:49.4,52.6)and72.7%.Conclusion:(1) p65、VEGF and MMP-9testing could predict the invasive behaviorand higher clinical Enneking stage of osteosarcoma;(2) MMP-9correlate with themetastasis behavior of osteosarcoma;(3) p65correlates with VEGF and MMP-9positively,and VEGF did not correlate with MMP-9;(4) p65and VEGF positive stainingfavors unfavorable prognosis.
