Objective:To explore the expression of vascular endothelial cell growth factor (VEGF) and bone morphogenetic protein-2(BMP-2) in osteosarcoma tissue and to analyze their association with clinical diagnosis, pathological grade, metastasis and prognosis of osteosarcoma.Methods: Immunohistochemical staining(SP method)was used to detect the expression of VEGF, BMP-2 and CD34 in specimens from 41 cases of osteosarcoma. To calculate microvessel density and overall analyze information of all the 41 cases.Result:In 41 cases,the positive rate of VEGF and BMP-2 were 68.2%(28/41)and 61.0%(25/41).The expression of VEGF and BMP-2 were not significantly correlated to tumor,s size,Price,s grade and Dahlin,s difierentiation(P>0.05);The positive rate of BMP-2(75%)and MVD (40.25±8.02)in VEGF positive patients were respectively much higher than BMP-2(30.8%)and MVD(15.54±5.87)in VEGF negative patients(P﹤0.05);The positive rate of VEGF(92.9%)and BMP-2(85.7%)in patients with metastasis were respectively remarkably higher than VEGF(55.6%)and BMP-2(48.1%)in patients without metastasis(P﹤0.05);The five-year survival rate in patients with positive expression of VEGF(7.1%)and BMP-2(4%)were significantly lower than that(46.2%and 43.8%)in negative control groug(P﹤0.05).Conclusion:High expression of VEGF and BMP-2 were found in osteosarcoma tissue.The expression of VEGF was closely correlated to the expression of BMP-2 and MVD.Detection of VEGF and BMP-2 expressions and MVD may be useful for judging metastasis and prognosis of osteosarcoma.
