The Significance And The Expression Of OPN And IL-1β In Osteoarthritic Articular Cartilage

Backgrounds:Osteoarthritis (OA) is the most common form of arthritis disease, OA is the most easily to involve the knee joint including over60more than about70%with different degrees of knee osteoarthritis, western developed countries joint prevalence rate is higher, and can be more than50%morbidity. With the improve-ment of national living standard in our country, an aging society, the incidence of osteoarthritis rising year by year, the related cytokines in osteoarthritis study beca-mea key research direction.Objective:By detecting osteopontin (OPN), interleukin1β(IL-1β) in bone arthritis with different expressions of articular cartilage degeneration discussnig OPN and IL-1β(3roles in OA in the process of occurrence and development.Methods:Methods:42cases of osteoarthritis cartilage specimens and10cases of normal knee articular ca r-tilage specimens are selected,eliminating infectious arthritis and rheumatoid arthritis(ra), having HE staining and red O staining after routine treatment,according to the modifie-d Mankin pathologic grades of the cartilage group:10cases of normal articular cartilage specimens,13cases of OA mild cartilage degeneration specimens,17cases of OA moder ate cartilage degeneration specimens,12cases of severe degeneration OA cartilage speci-mens. Usinimmunohistochemical methods to detect each group of cartilage in the expressi-on of OPN andIL-1β, half quantitative analysis using MI AS image analysis system by usi ng the software of SPSS17.0having statistical analysis to the result of the experiment.Results:HE staining:Bying compare the contrast group with the normal group,the contrast group all have the degeneration with chondrocytes and matrix.Immunohi stochemical:1, the OPN average grey value is:normal group is0.1627±0.0077, mild group is0.1812±0.0054, moderate group is0.2148±0.0241, severe0.2739±0.0059. All groups are compared that have statistical significance (P<0.05),modified Mankin OPN and pathologic grading r=0.872, were positively correlated.2, the IL-1βaverage grey value is:normal group is0.1659±0.0594, mild group is0.1836±0.0465,and moderate group is0.2363±0.0756,severe group is0.2786±0.1224. The comparison group has statistical significance (P<0.05),they arepositiveel y cone lated between the IL-1βand the improved Mankin pathological gradin-gr=0.942.Conclusion:OPN, IL-1β are highly expressed in cartilage of osteoarthritis, and associat ed with the onset of osteoarthritis and its progress.OPN and expression of IL-1β wa s positively correlated with the degree of degeneration of articular cartilage.
