Objective:To explore the effect of quercetin extracted from eucommia leaf onmesenchym stem cell (MSC) proliferation and its possible signal transductionmechanism.Methods:BMSC culture was performed with the adhering culture method.Usingthe phase contrast microscope to observe cell morohology and flow cytometry todetect the cell surface antigen to identify the BMSC.The third generation of BMSCwas used as experimental material.The experimental group was designed with0.5、1、2、4μg·mL-1of quercetin extracted from eucommia leaf.The group withoutquercetin was set as the control group.By using the MTT method to detect theproliferation of BMSC. Adding the end concentration0,0.5,1,2,4μg·mL-1of quercetinextracted from eucommia leaf to the BMSC for15min after starving the BMSC for6h by the serum-free and phenol red-free medium.Then we detemine the changes ofthe Phosphorylation of the ERK level using the Western blot.Results:Cell surface antigen detection is CD90(+)、CD29(+) and CD45(-).MTTresults showed that the quercetin extracted from eucommia leaf can promote BMSCprolieferation in a concentration dependent manner.Western blot results proved thisdrug can stimulate the Phosphorylation of the ERK.Conclusion:The Quercetin extracted from eucommia leaf Promote the prolifera-tion of bone marrow derived mesenchymal stem cells through the Phosphorylation ofthe ERK pathway.
