Objective:This subject, using Interstitial Cells of Cajal(ICC)as gastrointestinal pacemakercells, established the animal model of stress-induced spleen exhaustion damage toexplore the pathogenesis of gastrointestinal function disorder and research thetherapeutic effect, mechanism of action of classical prescription named BuZhongYiQiDecoction on gastrointestinal function disorder. This subject is based on variation ofICC such as structures, functions, gastrointestinal dynamics experiment, superfinemorphological observation and gene expression.Meanwhile, this thesis, obtained dynamic science information, explored theresearch status and development trend of this classical prescription-BuzhongYiqiDecoction from the perspective of literature metrology. Combining literature researchand experimental research, this thesisstrives to push the research on BuzhongYiqiDecoction into a broader, deeper and more comprehensive level to provide somescientific experimental data, new treatment strategies and some targets of drugsintervening action for its clinical application.Methods:(1) experimental studyUsing swimming exhaustion methods built spleen damage model of mice andrats, caused by exhaustion and stress, comparing the swimming time, general status,weight change, ability to resist fatigue of each group; using gastric emptying andintestinal propulsion experiment to observe the gastrointestinal dynamics change;cutting out the gastric antrum of rats, using Real Time quantitative Polymerase ChainReaction (Real-Time qPCR) method to detect the variation of gene expressionquantity of c-kit target and using transmission electron microscopy to observe the ICCultrastructure changes.(2) literature researchUsed the CQVIP science and technology periodical database as the mainsource of materials, chose research on BuzhongYiqi Decoction from1989to2013asthe search object, which applied analysis methods of discrete law of Bradford literature measurement to carry out the statistical analysis on five aspectsrespectively literature’s periodical distribution, literature type, disease research andpharmacological experiments to seek quantitative description in order to reveal thedistribution features and explore the development trend of the research on thisclassical prescription, then put forward suggestions to existing problems so as to helpfurther study on BuzhongYiqi Decoction in the future.Results:(1)experimental studyThe model of this experiment successfully simulated the exhaustion spleen ofstress-induced exhaustionmice and rats; the study indicated that animals of the modelgroup generally presented spleen deficiency, a slow increase of body weight, decreaseof the anti-fatigueability, gastrointestinal disorders; BuzhongYiQi decoction canimprove the above damaged changes caused by Laojuan and stress. Gene expressionof the distal c-kit of model rats showed the change trend from gradual increase ofirritabilityto traumatic decrease as the research went. Group of BuzhongYiqiDecoction in general showed a trend of restorative regulation to normal expression.Under the electron microscope, the injury of the ultrastructure of the ICC, appeared inmodel rats, couldbe effectively cured by the decoction.(2) literature researchInductive analysis of literature of BuzhongYiqi Decoction in25years’researchshows that the document number of both the current study and estimated researchisgenerally on the rise year by year. Literature periodical distribution statistical analysisshows that the contents are relatively concentrated with low literature dispersion, andthere has formed a main periodical group of research literature. The literature of theresearch mainly focuses on clinical application with internal diseases being the majorsubjects and constipation, splanchnic prolapse and fever the most commonly studieddiseases. Study of pharmacological experimental and literaturereview is relativelyfewer while the hot spot is the regulation of gastrointestinal function and immunefunction.Conclusion:(1) experimental studyBuzhongYiqi Decoction can regulate the spleen and stomach function, and curegastrointestinal dysfunction. This prescription may adjust expression quantity of the c-kit mRNA to regulate gastrointestinal function disorder, which may be themechanism of BuzhongYiqi Decoction’s treatment of gastrointestinal disorder. Thereis a close connection between the change of ICC’s ultrastructure and gastrointestinalfunction disorder, and BuzhongYiqi Decoction can improve these changes to a certainextent, that may be the another important mechanism of BuzhongYiqi Decoction’streatment of gastrointestinal dynamic conditions.(2) literature researchComprehensive analysis showed that clinically, BuzhongYiqi Decoctionwasmainly used to cure gastrointestinal diseases, and this purpose has been studied inpharmacological experiment, however, experiment study of its regulating stressreaction is relatively fewer. Nowadays, the incidence of psychosomatic disease causedby exhaustion and stress rises year by year, resulting a big proportion of abdominaldistention, diarrhea and other gastrointestinal dysfunction in digestive psychosomaticdiseases. Gastrointestinal function disorder and psychosomatic stress response is moreclosely related, therefore, it is necessary to carry out the research with BuzhongYiqiDecoction intervention of pharmacological effects and mechanism of action ofgastrointestinal function disorder diseases caused by psychosomatic stress. For furtherresearch, the following aspects can be taken into consideration: the first is tostrengthen basic research; second, to strengthen the pharmacological experiment ofBuzhongYiqi Decoction; third, to research with application of the multidisciplinaryknowledge and contemporary natural science, modern medicine and other subjects ofnew knowledge; fourth, to combine with literature, clinical, and experiment.
