| Pedicularis henryi Maxim. is a medicinal plant which belongs to the genusPedicularis L. of family Scrophulariaceae. Here, It was selected as the researchobject to study systematically on the pharmacognosy, chemical constituents, determination or content, acute toxicity and quality control. The research results are asfollows:1. Pharmacognosy of Pedicularis henryi Maxim. were studied combinedtraditional identification experience and modern scientific technologies, to evaluatequality of Pedicularis henryi Maxim. for its origin, micro-character andphysicochemical properties.2. To studies on chemical constituents from the roots of Pedicularis henryiMaxim.,10compounds were identified by phsicochemical properties and spectralanalysis. These compounds were identified as follows: Syringaresinolmono-β-D-glucoside(1), Plantarenaloside(2), Phenylacetic acid (3),2’’,3’’-Diacetylmartynoside(4), cis-Martynoside A (5), Martynoside (6),cis-Martynoside(7), Leucoseceptoside A (8), Jionoside D (9), Aucubin (10).They have been isolated from thisplant for the first time.3. Determination of aucubin in Pedicularis henryi Maxim. The content of Aucubinwere determined by HPLC. The content of Aucubin≥0.17%was estabished as thequantitative determination criteria. The method is simple, accurate with goodreproducibility and can be used for the quality control of roots of Pedicularis henryiMaxim.4. To investigate the toxicity effect, the acute toxicity test was carried out ongood health mice. The maximum concentration of ethanol extract from Pedicularishenryi Maxim. was0.85g/ml and the possible maximal dose volumes of mice was 0.4ml/10g. The acute toxicity test on mice with this dose showed that all mice wereall survival and there was no abnormal changes on the appearance, appetite, behavioral activity and excretion after14days intervention. These mice weresacrificed and dissected, the organs were as usual. It suggested that the maxiumdosage of mouse is34.00g/kg.bw, equivalent to234.15g/kg.bw of the crude drug. themaximum concentration of ethanol extract was468times of the dosage used in clinicfor an adult. The acute toxicity experiments show that Pedicularis henryi Maxim. issafe in the administration scope.5. To draft out the quality criteria protocol of Pedicularis henryi Maxim. Thestandards of water, extractives, total ash, acid-insoluble ash, heavy metals content andharmful elements were established: water≤16.0%, extractives≥23.0%, total ash≤6.0%, acid-insoluble ash≤1.9%.The heavy metals limited were Cu<20ppm.In this paper, through the studies mentioned above on the roots of Pedicularishenryi Maxim., it enriched the systematically pharmacognostic research, providedtheoretical proof and practical reference for developing the resource of medicinalplant in genus Pedicularis. |
