BackgroundPogejiuxin decoction is a famous clinical prescription to cure Cardio-vascular Disease created by a famous Chinese doctor named Li Ke in present era. The decoction is composed by Aconite Radix Laterlis, Glycyrrhizae Radix et Rhizoma, Zingiber officinal is, Cornus officinalis, Draconis,Streae Concha and so on, among which the most characteristics of this decoction is use large dose of Aconite and Cornus.For the treatment of heart failure, with a maximum dose of aconite almost to500g one day and single prescription dosage in the range of30-200g,exceed daily dose of aconite which prescribed by Chinese Pharmacopoeia of2010edition3-15g a lot. Although it proves to be significant effect on various types of severe heart failure, but high doses of toxic herbs-Aconite Radix Laterlis, limiting this prescription in clinical use and promotion.The Current research to this decoction is mainly limited to pharmacological mechanisms and clinical effects, studies on chemical composition of the decoction is almost blank, the compatibility factors influence on chemical composition is not yet concerned by scholars.Object ivePogejiuxin decoction are mainly to treat severe cardiovascular disease crisis for older people, large doses of aconite achieve "Yang deficiency" effect were emphasized in the clinical application. But the decoction is rarely showed adverse events in clinical practice, we assume that its toxic material basis can be controlled by compatcompound herbs in the progress of decoction.This paper plan to describe the change of toxic alkaloids in aconite during decoction process and investigate influence of different compatibility to the contents of three kinds of ester-alkaloids(benzoylmesaconine, benzoylaconine,benzoylhypaconine)in Pogejiuxin decoction,explaining about the compatibility rule for this decoction from the perspective of the material basis, thus provided information for the perspective of clinical safety, rational use of the decoction by certain chemical data support.Methods1. The content of six kinds of ester-alkaloids from aconite which processedin three different ways were determined by using highperformance liquid chromatography;Then established an HPLC method for determining three monoester-alkaloids in aconite decoction, monitoring the content of ester alkaloids with the dynamic changes of boiling time to understand the distribution of ester alkaloids in decoction.2. Established an method for determining monoester alkaloids for Pogejiuxin decoction, observe the quality changes of three kinds of monoester-alkaloids through different compatibility and different doses of Aconite.3. By measuring ester alkaloids from Aconite, quantitative research the laws of Aconite-Cornus under different compatible ratio influence on ester alkaloids.Resultsl.The main toxic ester alkaloids were diester-type alkaloids which most came from crude Aconite, but processed Aconite contained mainly were monoester-type alkaloids. It showed that different processing methods lead composition and content of ester-type alkaloids are quite different. Compared with crude aconite, the processed Aconite led to a huge loss of ester-type alkaloids in the process. In the three kinds of aconite decoction process, ester alkaloids mainly present as the form of mono-ester alkaloids, among which the content of the highest was benzoylmeaconine. In the process of boiling120min to300min, three kinds of momo-ester Aconitine was basically stable, with no significant degradation.2. Compared with aconitum decoct at the same volume water in single, the contents of mono-ester alkaloids of Pogejiuxin decoction and its compatibility of each group relatively had some decline, among them all-party compatibility group was the lowest mono-ester alkaloids and Cornus can significantly reduce ester-type alkaloids.Moreover, with the dose of aconite increased the contents of alkaloids of decotion growed at the same time,but its growth rate less than the rate of aconite.3. Compared with single Aconite, Aconitum-Cornus compated with different ratio could decrease ester-type alkaloids in dissolution rate significantly. We noticed that the dissolution rate relating to the proportion of the ratio, the greater ratio the lower dissolution rate was. Water-soluble components from Cornus may couse the results because those chemicals can make complexation reaction with ester-type alkaloids such reduced its concentration in the decoction.ConelusionWe find The Pogejiuxin decoction compat in different ways can influence ester-alkaloids in a regular way. Compared with the single aconite, the content of ester alkaloids from aconite changed after compatibility, and the dominant factor restricts ester alkaloid content is the Cornus to this decoction.Further research shows that the possible ways restricting ester alkaloids are water-soluble organic ingredients from Cornus which leads chemical reactions among them. Due to the content of efficacy material basis in prescription has changed by compatibility, resulting in decreased aconite toxicity, thus provides chemical data support for using large doses of Aconite in the clinical.
