Objective:To optimize the extract method of jujubosides from Semen Zizyphi Spinosae(SZS).Discuss fingerprint and serum fingerprint about compatibility alkaloids with Saponins from SZS.Through the establishment model of chronic stress,insomnia depression of mice,observe the effect of alkaloids and saponins from SZS on behaviors and monoamine neurotransmitters and antioxidant activity of mice with chronic stress insomnia depression.And discuss whether the alkaloids and saponins from SZS have depression-relieving and spirit-quieting effects.Method:1.Optimize the extract method of saponins from SZS.Use the high performance liquid chromatography-diode array detection(HPLC-DAD),study on alkaloids and saponins from SZS compatibility of fingerprint and serum fingerprint.All this work lay the foundation for the spectral correlation effect research in the future.2.We chose the chronic unpredictable stress animal model of depression to judge the sedative-hypnotic and antidepressant effects of alkaloids and saponins from SZS.The mechanisms were examined by measuring monoamine neurotransmitters in mice fraction and antioxidant status of brain.Result:1.The content of total sapoin obtained by the new method was more than 50%and had good solubility in water.It is advantageous to the experimentation on animal.The best dose of alkaloids and saponins were at the ratio of 1 to 1 and 1 to 10.In the serum fingerprint experiments,we speculate that the compatibility can promote zizyphusine into blood.2.In the mice tail suspension test and forced swimming test,SZS treatment groups compared with model group,mice TST and FST immobility time were significantly shorter,have statistically significant(P<0.05).Locomotor activity in mice experiments,the treatment groups compared with model group,have a significant increase in the number of standing and activity,have statistically significant(P<0.05).According to the result of NE?DA?5-HT content in hippocampus and cortex,the groups of alkaloids and saponins decreased significantly compared with normal group,have statistically significant(P<0.05).The weights increased were marked less than those of model group(P<0.05).All above account for that compatibility alkaloids with saponmns from SZS have sedative-hypnotic and antidepressant effects.Compatibility alkaloids with Saponins from SZS have obviouse synergy effects with pentobarbital sodium of under threshold dose and threshold dose lead to sedative and hypnotice on mice.3.Alkaloids and saponins from SZS compatibility significantly raised the activity of T-AOC,GSH-PX,CAT and decreased the content of NO.Conclusion:According to the speciality of jujubosides,the optimized extract method was established.The method is simple,feasible and suitable for industry production.By using HPLC-DAD,the fingerprint alkaloids and saponins from SZS compatibility was established.The serum fingerprint experiments establishes the foundation for spectral correlation studies and identifition of effective components.The pharmacodyna.ic result showed alkaloids and saponins from SZS produced an antidepressant-like effect in a dose-dependent manner.In addition,the mechanism of action involves the serotonergic and noradrenergic systems.The inhibition of lipid peroxidation and the scavening of free radical may account for the observed antidepressant effect.Experimental results show that the alkaloid and total saponins have exact synergy depression-relieving and spirit-quieting effects.
