Objective: Asthma is one of the most common chronic diseases in the world,chronic airway inflammation is its basic feature, present in all asthma patient. It isimportant for asthma control,reducing the number of asthma attacks and reducingasthma mortality that the accurate, real-time monitoring of airway inflammation inasthma patients.Periostin is an extracellular matrix protein which was originallydetected in osteblasts.It is found that Periostin plays an importmant role in thepathogenesis of asthma in recent studies,for example airway eosinophilia, airwayhyperresponsiveness and airway remodeling.This study was designed to investigate therole of Periostin in bronchial asthma and the relationship between Periostin and serumIgE, the percentage of eosinophils (EOS%), the forced expiration volume in one secondpercentage predicted(FEV1pred%), and to investigate the changes of peripheral bloodPeriostin, IgE and EOS%levels after treatment in acute asthma patients, and study onPeriostin’s efficacy for evaluation and prognosis of asthma and its clinical significance.Methods:Collecting47acute asthma patients hospitalized in the Second AffiliatedHospital of Dalian Medical University during2013June to2014March,age15-73yearsold, mean age (53.98±14.91) years old, consisting of20men and27women.Thepatients with bronchial asthma were classified into two groups which were the mildgroup (n=26) and the moderate-severe group (n=21) in the light of the order of severityof asthma. Meantime collecting19cases of healthy people as the normal control group,they were no smoking, no asthma, no allergic rhinitis and no allergy history, withoutdisease of circulatory system, digestive system, respiratory system,rheumatoid immunesystem and so on, no history of recent infection and drug history.All of the subjectswere determined by double antibody sandwich enzyme-linked immunosorbent assay (ELISA) for determination of serum Periostin and IgE levels, hematology analyzer wasused to examine EOS%,repeating pulmonary function test3times by lung functioninstrument, determinating FEV1pred%, selecting the best value. The value was recordedby group.In addition,47cases of acute asthma patients were relieved aftertreatment(asthma symptoms and signs were disappeared).When they weredischarged,reviewing the serum Periostin, IgE and EOS%levels of these methods andrecording. The data were analyzed by SPSS17.0statistical software,P<0.05wasconsidered statistically significant.Results:1. the serum level of Periostin in acute asthma were significantly higherthan that of control subjects respectively,the difference was statisticallysignificant(P<0.05).2.the serum level of Periostin in moderate-severe acute asthma werehigher than mild acute asthma,the difference was statisticallysignificant(P<0.05),however,there is no difference about IgE,EOS%between the mildgroup and the moderate-severe acute group.3.the serum level of Periostin in acute asthma patients who had beentreated was lower than those before therapy, the difference was statistically significant(P<0.05),however there is no difference about IgE,EOS%between untreated andtreated.4.the serum level Periostin in acute asthma patients who had been relivedwas lower than the normal control group, the difference was statistically significant(P<0.05).5.in acute asthma, there was positive correlation between the serumlevels of Periostin and IgE,EOS%(P<0.05),there was negative correlation between theserum levels of Periostin and FEV1pred%(P<0.05).Conclusion:Periostin is closely related to the pathogenesis,severity and prognosisof bronchial asthma.It may paticipate in the pathogenesis of asthma with IgE andeosinophils, and it is more sensitive than IgE and EOS%for providing reference indexfor the diagnosis,treatment response and prognosis of bronchial asthma.
