ObjectiveTo observe the intervention effects of Changhe decoction(CHD)to acuteradiation enteritis in rats,which introduced by medical X-ray.And explore infla-mmatory mediators nitric oxide (NO)、interleukin6(IL-6) levels,the antioxidantdefense system of superoxide dismutase (SOD)、glutathione peroxidase (GSH-Px)activity of intestinal tissue in rat models,which may affect with CHD.And disc-ussed the related mechanism of CHD for treatment ARE,supply experimentalevidence for CHD’s clinical treatment and experimental basis.MethodsForty six Sprague-Dawley rats were randomly divided into four goups.Therats in Goup1received CHD underwent irra-diation;The rats in Goup2recei-ved Compound Glutamin Entersoluble capsule(CGEC) underwent irra-diation;The rats in Goup3received0.9%Nacl underwent irradiation alone as model;The rats in Goup4received0.9%Nacl underwent no irradiation as normal con-trol.Rats in the all groups were exposed to a single dose of10Gy by6MVhigher energy X ray on the whole abdominal region,to establish the acute radia-tion enteritis models,except those normal control.Then all the rats were intraga-strically administered with corresponding agents for a consecutive of7days.Then8cm jejunum and ileum samples were obtained from each animal undergeneral anesthesia.The morphologic indices were examined by light microscopyand the image analysis system.The inflammatory mediators nitric oxide (NO)levels,Interleukin-6(IL-6) levels,and the antioxidant defense system of superoxi-de dismutase(SOD)and glutathione peroxidase(GSH-Px) activity in the smallintestinal homogenates,were measured by Multifunctional microplate reader.Results1.This experiment used6mv medical X-ray exposure to the whole abdom-en of rat, which successfully established the rat model of acute radioactive en- teritis(ARE).2.Groups of rats changed in general and weight:After irradiation,the group1and gropu2had good general conditions and defecation than the group3,especially from4th to7th day.In8th day,compared with group3,group1hadsignificantly increased body weight of rats(P<0.01).And there were not signifi-cantly different in weight between group1and group2(P>0.05).3.The morphologic indices changed in the small intestine:After irradiation,group1and group2of small intestinal mucosa were complete,the inflammationwas lighter,while group3appeared mucosal necrosis,fall off,gland atrophy,heavyinflammatory response,and so on.The group1had significantly higher villus hei-ght,deeper crypt,thicker mucosa and entire wall thickness than group3(P<0.05),and the above parameters were not significantly different between group1andgroup2(P>0.05).4.Biochemical indices:The group1and group2had significantly lower NOand IL-6levels in the intestine compared with the group3(P<0.01).Group1hadsignificantly lower IL-6levels with the group2(P<0.05),and there were not sig-ificantly different in NO levels between them(P>0.05).There were not signi-yantly different in NO and IL-6levels between group1and group4(P>0.05).Compared with group,group1and2had significantly more SOD and GSH-Pxactivity in the small intestine tissue (P<0.05).And there were not significantlydifferent in SOD and GSH-Px activity between group1and group2(P>0.05).Conclusion1.It was indicated that Changhe decoction(CHD) had protective effect agai-nst intestine mucosa damage in acute radiation enteritis rat model(ARE).2.Changhe decoction(CHD)can protect the ARE rat small intestine mucosas-mal, and rsignificantly reduce inflammation and tissue damage.3.Reducing the inflammatory mediators NO and IL-6levels,increasing theantioxidant defense system of superoxide dismutase(SOD)levels and glutathi-one peroxidase(GSH-Px) activity in the small intestinal homogenates were maybe one of the mechanisms for treatment of ARE.
