ObjectivesTo discuss the effect and mechanism of Liangxueyuchang Decoction onradiation-induced enteritis in a rat model caused by radiation of different doses.Through observation of the general condition and state of defecation, thepathology changes of jejunum and rectum under light microscope, thecalculation of crypt survival rate for jejunum,the score by Hovdenak for rectum,and using the method of bacterial culture for the detection of bacterialtranslocation rate for mesenteric lymph nodes,using the method of nitratereductase for the determination of NO levels of intestinal mucosa and using inkprogradation rate for the evaluation of gastrointestinal motility, the aim is toprovide experimental evidence for clinical research.Methods162SD rats were randomly divided into four groups. Group A(n=18) wasthe normal group, Group B(n=48)、Group C(n=48) and Group D(n=48) wererespectively underwent whole abdominal irradiation with X-rays at an singledose of5Gy、10Gy and15Gy, After the completion of radiation enteritis model,group B、group C and Group D were randomly divided into two groups, group B1、group C1and group D1after irradiation were administrated by i.g. withdistilled water, group B2、group C2and group D2after irradiation were withLiangxueyuchang Decoction. From the first day after irradiation, the groupswere consecutively administrated by i.g for7days. General condition and stateof defecation of the rats were observed every day. All groups were taken onethird rats for experiment on the2nd、4thand7thd after irradiation.Firstly, allgroups with irradiation were administrated by i.g. with experimental ink(including5%active carbon powder,10%Arabia gum) for ink progradation ratein order to evaluate gastrointestinal motility,after30minites,they were taken thejejunum, rectal tissue and mesenteric lymph nodes.The pathology changes ofjejunum under light microscope were observed,and the jejunum samples werecollected for crypt survival assay, the rectums were evaluated microscopicallyusing a scoring system reported by Hovdenak et al,bacterial translocation wasexamined,and the content of NO of the intestinal mucosa were evaluated.Results(1) The general condition and state of defecation of the LiangxueyuchangDecoction group have an advantage than the distilled water group afterradiation, especially after5-7days, except B group.(2) Jejunum、rectum mucous membrane of the Liangxueyuchang Decoctiongroup were near to complete and have an lighter inflammatory reactionsthan the distilled water group, especially B2、C2group,while there wereglandular atrophy, necrosis and abscission on the latter′S mucousmembrane,and B1group was not too bad.(3) The survival rate of gland were improved in the different extents on the4thand7thd for the Liangxueyuchang Decoction group in C、D group(P <0.05,P<0.01). (4) The scores of group D、group C and group B were higher than group A,thescores of the Liangxueyuchang Decoction group on the4thd is lower than ofthe distilled water group(P <0.05).(5) The condition of bacterial translocation after radiation exists in all groups,and the bacterial translocation rates of the Liangxueyuchang Decoctiongroup on the4thd is significantly lower than of the distilled water group(P<0.05, P <0.01).(6) The change of the content of NO of jejunum and rectum is basicallyconsistent.The content of NO of group C1was higher than group A on the4thand7thd (P <0.05), and the content of NO of the group whichadministrated by i.g.with Liangxueyuchang Decoction was lower obviouslythan the group with distilled water(P <0.05),But still not back to normal.(7) ink progradation rate for small intestine:the groups with irradiation were allhigher than group A, there was no significant difference between the groupwith Liangxueyuchang Decoction and the group with distilled water on the2ndd(P>0.05),and the group with Liangxueyuchang Decoction was lowerthan the group with distilled water on the4thand7thd(P<0.05),except groupB.ConclusionsLiangxueyuchang Decoction on acute radiation-induced enteritis iseffective, its efficacy is relevant to nuclear radiation dose and time ofadministration. Liangxueyuchang Decoction can accelerate recovery of injuredintestinal mucosal epithelium,reduce bacterial translocation and maintaingastrointestinal motility、normal structure and function of intestinal mucosa,its mechanism may be concerned with decreasing the NO concentration ofintestinal tract.
