Objective: Analyze the expressing level of miRNAs in CRC and probe into thepotentiality of miRNAs of being CRC diagnostic markers.and how the clinicalcharacteristics affect the expressing level of miRNA.Method:This study is try to analysis the expression level of1547miRNAs inCRC by QRT-PCR and the expressing level of miR-490-5p and miR-363in CRC withdifferent characteristics.Results: This study identified569miRNAs that were significantly dysregulatedin CRC relative to normal tissues (P<0.05) with paired-t test and107dysregulatedmiRNAs with nonpaired-t test (P<0.05). and the results indicate that they expressionare not identical from the statistic analysis of miR-490-5p and miR-363in CRC withdifferent characteristics. miR-490-5p and miR-363are down-regulated more than7times In stage II and6times stage III with P<0.05compared with normal tissues, instage IV the disregulation has no statistical significance with p>0.05even thoughdown-regulated more than2times. And then we performed ROC (receiver operatorcharacteristic curve) analysis the sensitivity and specificity, the consequence of ROCmiR-490-5p and miR-363can screen stage II-III CRC from normal person very wellwith AUC (area under the curve) over0.70and P<0.05.acorrding to stage IV, the result show that miR-490-5p and miR-363is not suitable be biomarker because ofAUC less than0.70or p>0.05. The expression level of miR-490-5p and miR-363descend more than5times with P<0.05in CRC with different genders, and ROCshow that miR-490-5p and miR-363can identify CRC from normal person.According to different pathology patients, the study indicates miR-490-5p andmiR-363`can distinguish CRC in middle differentiation not high differentiation fromnormal tissues. And then, we analysis their expression level in colon and rectal cancerrespectively. miR-490-5p and miR-363can act as biomarkers markers for rectal andcolon cancer except that miR-490-5p is not suitable for diagnosing rectal cancer fromnormal groups. At last, we mixed all samples together, and compare their expressionlevel with normal groups, we find that both of miR-490-5p and miR-363can screenCRC patients from normal person very well with AUC>0.70and P<0.05.Conclusion: There many miRNAs dysregulated in CRC,and the expressionlevel of miRNAs are not entirely consistent among different CRC with clinicalcharacteristic. MiR-490-5p and miR-363have the potentiality of being CRCdiagnostic markers.
