| Objectives Based on the high-throughput sequencing data of thyroid cancer in the The Cancer Genome Atlas,we analysis and filter the differentially expressed miRNAs in thyroid cancer.For the significantly differentially expressed miRNA-4709-3p and miRNA-34a-5p,the diagnostic value of their expression in serum for the diagnosis of papillary thyroid carcinoma will be explored,in order to provide a reference for the clinical optimization of early non-invasive diagnosis methods of papillary thyroid carcinoma.Methods Download the high-throughput miRNAs sequencing data of thyroid cancer from the TCGA database,analysis and filter out the miRNAs with significant differential expression and detect the expression in the sample.Collect fresh cancer and adjacent tissues from 30 PTC patients,and detect the expression of miRNA-4709-3p and miRNA-34a-5p in the tissues by fluorescence quantitative PCR,and collect 50 preoperative serum samples from benign and malignant patients and detect the expression of them.The histopathological results of surgery were used as the gold standard to determine the nature of thyroid nodules.Take the histopathological results of surgery as the gold standard to determine the nature of thyroid nodules.Multi-factor Logistic regression analysis was used to screen miRNAs and clinical characteristic variables which is closely related to diagnosis,analyze the sensitivity,specificity and Kappa value of serum miRNA-4709-3p and miRNA-34a-5p expression in the diagnosis of benign and malignant nodules,evaluate the clinical application value of serum miRNA-4709-3p and miRNA-34a-5p in the diagnosis of papillary thyroid carcinoma.Results 1 According to the sequencing data of miRNAs in TCGA,the differential expression of hsa-miR-4709-3p and hsa-miR-34a-5p between thyroid cancer tissue and normal tissue is the highest in thyroid cancer.2 The expression of miRNA-4709-3p and miRNA-34a-5p in PTC patients’ cancer and adjacent normal tissues is different.3 The expression of miRNA-4709-3p and miRNA-34a-5p in the serum samples of PTC patients and patients with benign thyroid nodules is different;The relative expression of the serum levels of miRNA-4709-3p and miRNA-34a-5p is closely related to the judgment of benign and malignant thyroid nodules.4 The area under the curve(AUC)of serum miRNA-34a-5p for the diagnosis of PTC is 0.910(95%CI: 0.837-0.958).When the relative expression level is 1.765 as the diagnostic threshold,the sensitivity is 92.0%,the specificity is 86.0%,and the Kappa value 0.78;The area under the curve(AUC)for the diagnosis of serum miRNA-4709-3 is 0.924(95%CI: 0.853-0.976).When the relative expression level is 2.395 as the diagnostic threshold,the sensitivity is 92.0%,the specificity is 80.0%,and the Kappa value 0.7.Conclusions The expression of serum miRNA-4709-3p and miRNA-34a-5p in patients with papillary thyroid carcinoma is significantly higher than that in benign nodules,which is helpful for the differential diagnosis of benign and malignant thyroid nodules.Them are expected to become papillary thyroid carcinoma early,non-invasive diagnostic markers Figure4;Table22;Reference 151... |
