Objective:To investigate the role of OPN in the formation of idiopathic calciumoxalate stone (ICS) by detecting the change in OPN mRNA using real-time RT-PCRand OPN protein using Western blot in the renal tissue of patients with ICS, and thechange in the concentration of OPN in ICS.Methods:From January2013to January2014,34cases of no-function kidneyswere collected after the nephrectomy in our hospital as the case group, including16cases of males,18cases of females, with an average age of50.7(range13to73)years. At the same time,38cases of normal renal tissue away from the tumor morethan3cm in the renal carcinoma were collected as the control group, including16cases of males and22cases of females, with an average age of50.1(range40to77)years. The expression of OPN mRNA in the tissues of kidney in the two groups wasdetected by Real-time RT-PCR and the expression of OPN protein in the two groupswas detected using Western blot. The concentration of OPN in the urine of thepatients in the two groups and the normal people was measured by enzyme linkedimmunosorbent assay (ELISA). The Data were expressed as the mean±SD andanalyzed using SPSS14.0statistical software (SPSS, Chicago, IL, USA).Comparisons among groups were performed using a two-sample t-test or analysis ofvariance.Results:The ΔCT of OPN mRNA in the renal tissues of the case and controlgroups were2.04±1.20and2.59±0.91, respectively. The difference was statisticallysignificant(P <0.05).The levels of the relative expression of OPN protein in therenal tissues of the case and control groups were0.776±0.071and0.449±0.085,respectively. The difference was statistically significan(tP <0.05). The concentrationof OPN in the urine of the patients in the case and control groups and the normalpeople were1.052±0.229,1.579±0.256, and1.498±0.155The differencebetween case and control group, and case and normal people were both statisticallysignificant(P <0.05). Conclusions: The higher expression of OPN mRNA and OPN protein in therenal tissue and the lower concentration of OPN in the urine in the patients with ICSplayed an important role in the formation of ICS.
