Objective:To investigate the effect of Ca2+on BMP2and MSX2expression and its relationship with GHS rat renal interstitial calcification.Methods:Extracted GHS renal tubular epithelial cells and divided into5groups randomly. Group1-4as experimental groups, group5as the control group. Group1-4add0.5mmol/L Ca2+,1.0mmol/L Ca2+,1.5mmol/L Ca2+and2.0mmol/L Ca2+respectively. Group5add PBS. Real-time fluorescent quantitative PCR and Western blotting test BMP2and MSX2relative expression of mRNA and protein between each experimental group and control group at12h,24h,48h respectively.Results:The BMP2and MSX2relative expression of mRNA and protein of each experimental group were significantly higher than control group at12h,24h,48h when Ca2+stimulates the GHS rat renal tubular epithelial cells(P<0.05). Each experimental group of BMP2and MSX2relative expression of mRNA and protein are reach peak when add1.5mmol/L Ca2+(P<0.05). With the increase of Ca2+concentration, all the experimental groups of BMP2and MSX2relative expression of mRNA and protein were increased gradually before reach the peak(P<0.05); Then, the both relative expression of mRNA and protein were reduced with Ca2+concentration increased(P<0.05).Conclusions:Ca2+stimulates GHS rat renal tubular epithelial cells highly express BMP2and MSX2,which may reveals high calcium activate the BMP-2-MSX2signaling pathways and participate in GHS rat renal interstitial calcification.
