Objective: From the experiment, investigate the mechanism of Xuebijing injection onliver transplantation ischemia reperfusion injury of ischemia and further research onreperfusion injury of liver transplantation, and master the technology related livertransplantation.Methods:(1)25Healthy Bama miniature pigs,were randomly divided into shamoperation group, control group and experimental group.(2) The experiment group wastreated with Xuebijing injection+UW solution: continuous low-pressure perfusion for20minutes; while the control group with UW liquid low-pressure perfusion without Xuebijinginjection in20minutes. The liver of sham operation group and liver transplantation of free,no ligation and blocking, not for liver resection.(3) Detection of animal survival rate andoperation time, anhepatic phase. Use the BeckmanCX7automatic biochemical analyzer forpreoperative and postoperative1,2,3,7days of liver function tests: ALT, AST, TBIL.(4)Using the BARD tissue biopsy needle liver biopsy, stained with HE, to observe thepathological changes in liver tissue with light microscope.(5) Detecting cell apoptosis byTUNE.(6)SOD: detected by xanthine oxidase method.(7)The ratio of liver tissueBax/Bcl-xl was detected by immune histochemical method.(8)Each group of data withmean±standard deviation said(x±s), using the SPSS18.0statistical software package forthe analysis of the data, using repeated measures analysis of variance, and then makecomparison between the two groups, with statistical significance in(P<0.05).Results:(1) The anhepatic phase and the operation time no differences between theexperimental group and the control group(P>0.05).(2) Before operation, experimentalgroup, control group without obvious difference of each index group and sham operationgroup(P>0.05).(3) Transplantation:1,2,3,7day of the experiment group and the controlgroup, the serum ALT, AST, TBIL levels were increased significantly, the experimentalgroup liver function values than the control group is low, compared with each other(P<0.05).(4) In liver tissues were observed under the light microscope, the control groupwith post transplant time prolonged liver pathological changes aggravated gradually, cell ofliver lobules obvious edema, a balloon like change of liver macrophage phagocytosis, cells increased, active, and easy to see the point, focal necrosis, portal area expandedsignificantly, edema, inflammation in the presence of mononuclear cells; liver tissue inexperimental group than in the control group with light, prolonged recovery; no significantchanges in liver tissue of sham operation group.(5) Detection of aoptotic hepatocytesTUNEL method: sham operation group liver tissue apoptosis at leAST, three groups ofmutual comparison (P<0.05).(6) SOD in the experimental group compared with thecontrol group(P<0.05).(7) The experimental group Bax/Bcl-xl was lower than the controlgroup (P<0.05).Conclusion:1、Xuebijing by enhancing the activity of SOD, reduce inflammation,reduce liver function values, has a protective effect on the liver.2、Xuebijing can regulatethe ratio of Bax、Bcl-xl, cell apoptosis, relieve the pathological changes, so as to protect theliver tissue.
