Objective Establish a stable fibrin gel release system and investigate the effects of different concentrations of thrombin, trasylol on the release rate of this system in vitro, simultaneously discuss the effect of growth factors and stability of its Biological activity on this system.Methods Establish different concentrations of thrombin, trasylol fibrin gel release system with fibrinogen, thrombin, trasylol, calcium chloride. Divide thrombin into three groups of group A (100IU/ml)、group B (150IU/ml)、group C(200IU/ml), and trasylol into groupl (0.14IU/ml)、 group2(0.35IU/ml)、group3(0.70IU/ml) in the same method. Observe the dissolution of each group in37℃water bath. To study the influence of the presence of growth factors on this fibrin gel release system, add four growth factors (PDGF-AB:200ug/L、VEGF:2ug/L、EGF:15ug/L、TGF-β1:15ug/L) in this system as the experimental group, non growth factors as the control group, observe the dissolution of these two group in37℃water bath. By Elisa assay to detect the concentration of growth factors in experimental group to know the stability of these growth factors’ biological activity in this release system.Results The volume of daily dissolved and the dissolution time in each group is different, when group A(1)、group A(2)、group A (3) lasted for4、5and6days respectively, group B(1)、group B(2)、group B(3) lasted for4、6and7days, and group C(1)、group C(2)、group C(3) lasted for3、6and5days. Through repeated-measures analysis of variance, the data above reach a result:the higher the concentration of trasylol, the slower dissolution rate of the FG release system, when thrombin at the sameconcentration(P<0.05), on the contrary, when trasylol at the sameconcentration, the higher the concentration of thrombin, the fasterdissolution rate of this system(P<0.05). Whereas, there is no effect onthe presence of growth factors on the dissolution of the FG releasesystem(P>0.05), experimental group and control group both lasted for9days in the same time, Furthermore, no significant difference was foundin the concentration of growth factors daily dissolution(P>0.05), thebiological activity of growth factors remains stable.Conclusion Fibrin gel delivery system can be used as a variety of growthfactor complex carrier release characteristics of variousconcentrations of the thrombin and aprotinin have different effects onthe system:The fibrin glue release system establish in vitro withfibrinogen, thrombin, trasylol, calcium chloride, can dissolve steady,slow and well-distributed. Under the condition of high concentrationsof thrombin, the increase of concentration of trasylol can acceleratethe dissolution rate of FG release system. In the different way, in thecase of a at low concentrations of trasylol, the increases concentrationof thrombin can delay the dissolution rate of this system. In addition,growth factors do not affect the dissolution of FG release system,andcan maintain stable biological activity.
