Objective: To repair full-thickness articular cartilage defects with chondrocyte/EGF/fibrin glue matrix. Method: I .To isolate the chondrocyte from rabbit joint cartilage and then to culture; 2.To study the effect of EGF to chondtocyte growth curve; 3.To prepare fibnn glue from the blood of rabbit through the method of ammonium sulfate precipitation; 4.To culture chondrocyte/EGF/fibrin glue in vitro, and to study the behavior of chongdrocyte in fibrin glue; 5. To repair full-thickness articular cartilage defects of rabbit with chondrocyte/EGF/fibrin glue matrix in which the chondrocyte density was 2.5x1 061m1. The articular cartilage defects were divided into three groups (group A, B and control) according to the chondrocyte used (autologous, allogeneic or no chondrocyte). The repair of defects was examined grossly, histologically and immune biochemically 1, 2, 3 months after operation respectively. Result: The quantity of the chondrocyte released from 45 minutes to 90 minutes was the largest; More chondrocyte was stained by trypan blue with the digestion time went on; The average cloning rate of chondrocyte was 29.4% in primary culture; 2.
