Objective:To investigate the effects on angiogenesis and functional outcome of exogenous vascular endothelial growth factor(VEGF) mixed with fibrin glue (FG) as a carrier repairing rat sciatic nerve injury.Methods:The rat sciatic nerve injury model was made by modulated Allen's weight drop method.36 wistar rats were randomly into experimental group(VEGF mixed with fibin glue group) and control group(gene transfection group),with 18 in each.Microvascular count(MVC),sciatic-nerve function index(SFI),motor nerve conduction velocity(MNCV),gross appearance,light microscopic observation were performed at 4,8,12 weeks after medication.Nerve sectional image analysis was performed at 8 and 12 weeks after medication to measure the mean diameter of neuraxis,thickness of myelin sheath and total number of myelinated fiber.Results:There was statistical difference in SFI,MVCV,the MVC of the nerve at 4,8 weeks(p<0.05) after medication and no statistical difference at 12week(p>0.05) compared with the control group.There was significant deviation in the mean diameter of neuraxis,thickness of myelin sheath and total number of myelinated fiber at 8 week after medication compared with the control group(p<0.05) and no significant deviation at 12 week(p>0.05).Conclusions:Fibrin glue can be used as a carrier for VEGF to repair injured sciatic nerves.These results indicate locally VEGF mixed with fibin glue can exert angiogenesis in the injured sciatic nerve area and enhance neurological function recovery of the sciatic nerve injured rats.There was no significant deviation compared with the administration of gene transfection.
