Objective: By copying the model of drug-induced liver injury, we explore promotingHuoxuehuazhuo capsule on acetaminophen-induced acute liver injury protectiveeffect and its possible mechanism.Method:40mice were randomly divided into four groups:normal control group,model group, DDB group, Huoxuehuazhuo capsule group, each with10mice. Theywere taken by intragastric administration after adaptive feeding7days, the druggroups were given the amount of the drug, the normal control group and model groupwere given normal saline, which lasted for7days. After intragastric administration,gave mice to the pellet feed and water. After model group, DDB group,Huoxuehuazhuo capsule group on days3,5,7being administered for6hours, theywere intraperitoneal injection APAP0.4g/kg (dissolved in40℃saline) for copyingliver injury model (modeling before8hours fasting and not but water); normalcontrol group was given normal saline by intraperitoneal injection, we begancollecting specimens after16hours in last modeling, and made liver tissue biopsy.Results: Compared with normal control group, activity of serum ALT in model groupwere augmented, yet concentration of GSH in liver tissue decreased with significantdifference (P<0.05), showing that models in mice were successful; Compared withmodel group, levels of serum ALT were reduced and concentration of liver tissueGSH were elevated in DDB group and Huoxuehuazhuo capsule group, withsignificant difference (P<0.05), indicating that Huoxuehuazhuo capsules and DDBcan improve levels of serum ALT and concentration of liver tissue GSH, to someextent, to protect liver cell; there was no significant difference betweenHuoxuehuazhuo capsules and DDB group, suggesting that Huoxuehuazhuo capsulesand DDB played a considerable role in treatment of acute liver injury. HE sectionsillustrated: model mice liver tissue had points, necrosis putrescence, infiltrationsurrounded by inflammatory cell, the cells slight swelling; each treatment grouppathological changes reduced. Conclusion: Huoxuehuazhuo capsules can reduce the activities of serum ALT, andraise the content of GSH in the liver tissue, to varying extent, to protect liver cell;meanwhile Huoxuehuazhuo capsules can ameliorate liver pathological changes, andimprove acetaminophen-induced liver cell necrosis and decrease infiltration byinflammatory cell in mice.
