Based on our previous research on Flos chrysanthemum indicium,this subject carried out more research on Yejuhuabaogan capsule(CBC),which was extracted and prepared from Flos chrysanthemum indicium by ourselves.We have been proceeded of pharmacodynamics investigation and screened out the drug concentration of anti-HBV and non-toxic;Acute toxicity test was performed in KM mice by the maximum dosage to evaluate the safety of CBC.Meanwhile,The CBC was tested for liver protective effect against mice liver injury induced by both alcohol and Con-A.HepG 2.2.15 cells was choosed to estimate the anti-HBV effect,the method was typical cell model of screening anti-HBV in vitro.The CCK-8 assay was used to examine HepG 2.2.15 cells' viability and hence find out the TC50 and TC0.On this basis,ELISA analysis method was used to detect the secretion of HBsAg and HBeAg by HepG2.2.15 cells on the sixth and ninth day after treatment with CBC,respectively.The results showed that CBC had inhibitory effect to the HepG 2.2.15 cells proliferation,and the inhibitory effect decreased with the decreased of the CBC concentration.The TC50 was 5.014 ?g/ml.Moreover,the secretion of HBsAg and HBeAg by HepG 2.2.15 cells were inhibited by CBC,and the inhibitory effect of HBeAg was more powerful than HBsAg and the sixth day was more powerful than the ninth day.The results showed that CBC has the effect of anti-HBV.In acute toxicity study,we used the method of the maximum dosage(0.6089 g/ml,40 ml/kg)to evaluate the safety of CBC,with continuous observation for 14 days.No death or abnormal clinical signs were found.The maximum dosage was 243 times higher than that of man's normal dose(0.1 g/kg).In order to study the liver protection effect,liver injury models were prepared with alcohol and Con-A.The former was chemical hepatic injury,and the latter was immune liver injury.Using Reitman-Frankel assay,TBA assay,WST-1 assay and ELISA assay,we detected the change of ALT,AST in serum,MDA,SOD in liver and TNF-? in serum.In addition,liver tissue pathology were observed after dissection.The protective effect of CBC against liver injury and the mechanism were overall evaluate.The results showed that CBC could reduce the ALT,AST,TNF-? in serum,the content of MDA in liver and increased the activity of SOD in liver of mice liver injury induced by alcohol and Con-A.Meanwhile,compared with the model group,CBC could improve the pathological change of liver,indicated that CBC could improve the acute liver injury induced by alcohol and Con-A.To sum up,it was the first time that the effects of CBC against HBV and protect liver injury and the mechanism induced dy alcohol and Con-A were confirmed,furthermore,some statistical analysis were finshed on the relationship between the dose of CBC and the liver protective effect.I hope this paper can provide some scientific reference value for anti-HBV and liver protection dural role of traditional Chinese medicine and lay the foundation for the research and development of health care medicine.
