ObjestiveTo investigate the genetic susceptibility correlationship of transcription factor7-like2(TCF7L2) gene rs7903146C/T、rs290487C/T、rs11196205G/C、rs12255372G/T polymorphism and Chinese Han population in Jiangxi Province with gestationaldiabetes mellitus.MethodProcess of object:Information was collected retrospectively from100Pregnantwomen with gestational diabetes Mellitus had been diagnosed and100healthyPregnant women who delivered at jiangxi Women and Children Hospital and HealthInstitute during January2010and June2013.Experimental method:By adopting the method of case-control study,The clinicalindicators and glucose metabolism related indicators were measured.Genomic DNAwas extracted from peripheral blood with Blood genomic DNA extraction kitextraction method.Allele-specific PCR was used to analyze the SNPs of TCF7L2rs7903146、rs290487、rs11196205、rs12255372.Statistic analysisTCF7L2gene rs7903146, rs290487, rs11196205, rs12255372genotypedistributions were the Hardy-Weinberg equilibrium (P>0.05).Statistical analysis wasconducted with Pearson chi-square test and t-test (SPSS17.0software) and P <0.05was considered statistically significant.ogistic regression analysis was an independentrisk factor binomial classification variables to P <0.05was considered statisticallysignificant.Results1The distribution frequencies of genotypes CC,CT and TT of TCF7L2geners7903146position in Jiangxi pregnancies were47.5%、37.0%and15.5%;Thefrequency of CC,CT and TT genotype of TCF7L2rs290487position were14.0%、35.0%and51.0%;And the frequency of GG and CC genotype of TCF7L2rs11196205position were99,5%、0.5%;And the frequency of GG genotype of TCF7L2rs12255372position were100%.2TCF7L2gene rs7903146、rs290487、rs11196205、rs12255372polymorphismwas consistent with Hardy-Weinberg equilibrium law. the difference of distributiongenotype distribution and C/T allele frequency of rs7903146between the two groupswas statistically significan(tP<0.05).Compared with C allele carriers,TT genotypecarriers had a4.195fold increased risk of GDM (95%CI1.715~10.266);P=0.002.No differences of genotype and allelic frequencies at rs290487、rs11196205and rs12255372were observed between the cases and the contrlis.3BMI, FPG, FINS, HOMA-IR between the case and control groups werestatistically significant (P <0.05).4Logistic regression analysis showed after adjusting for age,gestationalage,BMI, FPG, FINS, and neonatal birth weight, pregnant women with TT genotypecarriers of rs7903146were more prone to hyperglycemia compared with the C allelecarriers(OR2.77,95%CI1.027~7.474);P=0.044.BMI, FPG and FINS areindependent risk factors for GDM.Conclusions:The polymorphisms of rs7903146in TCF7L2gene may be associated withgestational diabetes mellitus in Jiangxi Han population; BMI, FPG, and FINS areindependent risk factors for GDM.
