Objective: To investigate the association between SNP allelic frequency of TNF-αand patients with gestational diabetes mellitus and their fetus.Methods: Fifty normal-pregnant women and fifty patients with gestational diabetesmellitus were from Beijing Aerospace General Hospital during March2012and December2013. The peripheral venous blood with mothers and cord blood were collected, and DNAwere extracted. Genotype frequencies analysis of restriction enzyme-857C/T、-863A/Cand-1031C/T fragment length were determined by polymerase chain reaction (PCR), andconfirmed by sequencing. Chi-square test was applied to analyze the Hardy-Weinbergequilibrium in genotype distribution. Data were analyzed t test and chi-square test.Results: The genotype distribution and allele frequency of-857C/T in TNF-αbetween GDM was significant difference to the normal control group. The proportion ofCT genotype and the frequency of allele genotype T in GDM group were significant morethan normal pregnant (P<0.05). There was no significance between the normal pregnantwomen and patients with GDM in the genotype distribution and allele frequency of-1031C/T and-863A/C locus (P>0.05). Compare the proportion of CC genotype in GDM groupand control with their fetus’ genotype with CT, had significant differences (P<0.05).Infetus of two group, we observed that the genotype distribution and allele frequency of-857C/T、-863A/C and-1031C/T in TNF-α in GDM were significant disparity to thenormal group. The genotype distribution and allele frequency of-857C/T in TNF-α inGDM were significant difference to the normal control group. The proportion of CTgenotype and the frequency of allele genotype T in GDM group was significant more thanthe control (P<0.05). The genotype distribution and allele frequency of-863A/C in TNF-αin GDM was significant difference to the normal control group. The proportion of CA genotype and the frequency of allele genotype A in GDM group were significant more thanthe control (P<0.05). The genotype distribution and allele frequency of-1031C/T in TNF-αin GDM was significant difference to the normal control group. The proportion of CCgenotype and the frequency of allele genotype C in GDM group was significant more thanthe control (P<0.05).Conclusion:1.The-857C'T polymorphism of TNF-α promoter gene may beassociated with GDM, whereas-1031C/T and-863A/C polymorphism have no relevantwith GDM.2. Three SNPS of TNF-α in fetus may increase the risk of mother withgestational diabetes.
