Hepatocellular carcinoma(HCC) is one of the common digestive mallignant tumors inChina, which has quick progress and poor prognosis. The feature that HCC is easy tometastasis affects the prognosis a lot, which makes the research about its metastasismechanism a hot issue. Betacellulin(BTC) is a member of EGF(epidermal growthfactor) family. Recent study shows it participates the progenesis and progression oftumors. This experiment is aimed at the expression difference of BTC in HCC cell linesand tissue and its effect on HCC.ObjectiveTo clarify the BTC expression difference between hepatoma cell and HCC cell lines,HCC tissue and tissue around HCC,to clarify its effect on HCC and mechnism.Methods1. Measure the expression difference of BTC in HCC cell lines(Huh7、HepG2、SMMC-7721and MHCC97H) and hepatoma cell line(HL-7702) by RT-PCR and Western-blot.2. Investigate the BTC protein expression difference in hepatocellular carcinoma tissueand tissue around the lesion by immunohistochemistry, then to analyze its relationshipwith pathological feature.3. For SMCC-7721and MHCC97-H cell lines that dowanreulate BTC with siRNA andstimulate with BTC, test their proliferation ablity with MTT, test their migration ablityand invasion ablity with transwell. Thus to analyse the effect of BTC on HCC cell lineand the mechanism.Results1. The expression of BTC in HCC cell lines is higher than it in hepatoma cell line, and theexpression of BTC increases with the increase of invasion ablitiy of HCC cell lines(P<0.05).2. BTC expression in HCC tissue is higher than it in tissue around the lesion, and itsexpression is higher in the lesion whose diameter is bigger than3cm and which showscapsular invasion(P<0.05).3. Downregulating BTC with siRNA decreased proliferation, migration and invation ablityof SMCC-7721and MHCC97-H, while stimulating SMCC-7721and MHCC97-Hwith BTC increased these ablities, through which is the BTC-PI3K/AKT-XIAP axis.ConclusionThe expression of BTC is higher in the HCC cell lines and the HCC tissue, whichaffect the pathological feature of HCC. And it can regulate the proliferation, migration andinvation abilty of HCC cells through the BTC-PI3K/AKT-XIAP axis...
