Objective: To investigate the expression of miR-23 a and XIAP in human hepatocellular carcinoma(HCC),the relationship between miR-23 a and XIAP,and the relationship between miR-23 a and XIAP in clinicopathological parameters of HCC patients.Methods: 1.The microarray was established and the changes of miR-23 a in 150 cases of human HCC and 136 cases corresponding to adjacent tissues were detected by in situ hybridization.2.The tissue microarray was established and the expression level of XIAP and Ki-67 in 150 cases of human HCC and 136 cases corresponding to adjacent tissues were detected by immunohistochemistry.Results: 1.The expression of miR-23 a and XIAP in HCC was localized in HCC cytoplasm and stained brownish yellow.2.The high expression rates of miR-23 a were 48.7%(73/150)and 77.2%(105/136)in HCC tissues and adjacent non-cancerous tissues respectively,there was significant difference between the two groups(P=0.001).The high expression rates of XIAP were 25.3%(38/150)and 22.1%(30/136)in HCC tissues and adjacent non-cancerous tissues respectively,there was no significant difference between the two groups;The expression rate of miR-23 a was higher in male than in female(P=0.037),HBs Ag positive was higher than that of HBs Ag negative patients(P = 0.022),large hepatocellular carcinoma was higher than that of small hepatocellular carcinoma(P = 0.021),and stage?-?was higher than that of stage?-?(P=0.002).While the high expression of XIAP protein in patients with small hepatocellular carcinoma is higher than that of large hepatocellular carcinoma(P=0.001),the?-?stage is higher than ?-?stage(P=0.009).In addition,both of them had no correlation with age,liver cirrhosis and histological grading.The expression of miR-23 a and XIAP protein in hepatocellular carcinoma was negatively correlated by Pearson's correlation(rs=-0.308,P<0.01).3.Pearson correlation test showed that miR-23 a and XIAP were significantly negatively correlated with the expression of XIAP(rs =-0.308,P<0.01).In HCC,miR-23 a could negatively control the expression of XIAP.Conclusion: 1.There was no significant difference in the expression of XIAP in HCC and corresponding adjacent tissues.2.In the HCC tissue,miR-23 a and XIAP were negatively correlated,miR-23 a may play a role in tumor suppressor genein HCC.3.miR-23 a and XIAP are expected to be targets for HCC diagnosis and treatment.
