PURPOSE: Tumor banks have the primary responsibility for collecting, storing anddisseminating samples of tissues and fluids, which are used by researchers to identifymolecular markers, prognostic indicators and therapeutic targets. The objective of this reviewwas to establish a colon tumour bank and describe four major areas as follows:(a) fund fortissue bank;(b)standard operation of specimen handling, processing and storage;(c)collectionof clinical data;(d) quality control of bank.MATERIALS AND METHODS: We approved of informed consent by Ethical Committeeof Hospital and establish fund and standard protocols for obtaining and storing samples.Colontumor tissues were collected from the patients of the affiliated hospital of NingXia MedicalUniversity, and we obtained clinical outcomes.Tissues snap-frozen in liquid nitrogen within30min after surgery then store at-80Freezer.We tested the quality of randomly selectedspecimens after one year, assessed the molecular integrity of specimens by DNA and RNAelectrophoresis, hematoxylin staining of frozen tissue sections were reviewed to assess thepercent tumour cell and reviewed clinical dates of stored tissues.RESULTS: We developed a protocol for the collection, storage, conservation and acesstumor samples. During a period of one year we stored587samples of tumor, peritumoral andnormal colon tissue of colon cancer patient, and549serum samples.RNA and DNAelectrophoresis showed RNA(assessed by visual estimate of the18s and28s ribosomal RNAbands by ethidium bromide staining) and the genomic DNA was of good quality performed oncollected samples.The percent tumour cell of tumour sample greater than70%and thespecimens have integrity clinical dates.CONCLUSIONS: Assessment shows availability and quality of collected colon tissue ofour tissue bank that are reliability and associated clinical data facilitating advances in molecular biology research.The successful established of tumour banking concerns include:protecting patient confidentiality, establishing standard operation, departments collaborationand strict quality control.
