Chuanshanlong is the rootstem of Dioscorea nipponica Makino and is bitter in taste and slightly cold in nature. It possesses the efficacy for curing rheumatic arthralgia ,chronic trachitis ,indigestion , subcutaneous swelling , malaria , internal injury due to overstain and Spain etc..The quality and effected factors of chuanshanlong cultivated by standard operating procedure was studied in this article. The evaluation method of quality and general technique used in the cultivation of chuanshanlong was established .The contents of diosgenin and total sapogenin were taken as the index and were determined by HPLC and colorimetry respectively . The samples cultivated in different cultural measure , cultivated in different grade ,collected in different producing area, different growth year and harvest time and processed in different method were determined in this article.The determined results and statistical analysis results of the samples cultivated in different density showed the contents of diosgenin and total sapogenin were highest in the sample cultivated in density of 67 × 19cm~2, and the statistical analysis results were best, so the optimal density was 67 × 19cm~2;the determined results and statistical analysis results of the samples cultivated in different height of shelf showed that the contents of diosgenin and total sapogenin were highest in the sample cultivated in shelf of 1.8m, and the statistical analysis results were best,the determined results and statistical analysis results of the sample cultivated with intercrop showed that the contents of diosgenin and total sapogenin were lower than the sample cultivated without intercrop, and the statistical analysis results were better, so the optimal height of shelf was 1.8m and the intercrop shouldn't be cultivated.
