Objective: Discussing about the relationship between human melatonin receptor genemutation and Han Chinese type2diabetes mellitus (T2DM).Method: Indagate the clinical case by contrasting295cases of T2DM patients of HanChinese in Ningxia with239cases of normal person. There are295cases of T2DM patients(100males and90females), Demographic index of acquisition cases (name, gender, age),common states(height, weight),medical history situation(the past medical history and familyhistory of T2DM),limosis biochemical indicator of blood (blood sugar, blood fat, trioxypurine)and blood pressure value. Blood pressure value was measured by mercurysphygmomanometer, Biochemical indicator of blood was measured by automatic biochemicalanalyzer of “OLIMPUSAU5400” type in Ningxia medical university second affiliatedhospital biochemical lab. The past medical history and family history of T2DM provided bycases. Data entry was finished together by my-self and my colleagues in the laboratory.239cases of matched group(male119,female120), limosis blood sugar and blood pressure ofmatched group are in normal range. The matched group came from the medical center ofsecond affiliated hospital of Ningxia medical university. All the patients are accord with thediabetes diagnosis standard of WHO in1999. Each individual were no blood relation.Measure the level of blood serum, blood fat and blood sugar. Analyse the relativity betweenpolymorphism of MTNR1B gene rs1387153site of Han Chinese in Ningxia and Type2diabates incidence with PCR-RFLP. Analyse the relation between age, gender, hypertension,blood fat, weight index, etc and polymorhphism of MTNR1B gene rs1387153site, type2 diabates incidence.Result: there are no differences in gender and age between two teams when they arecompared through statistics (P>0.05); The BMI, SBP, DBP, WAST, AIP in the team of T2DMis evidently higher than the other team in contrast(P<0.05). Biochemical indicator of blood:There are differences in statistics between two teams when it refers to FPG, TC, LDL, TG.However, they are the same in HDL (P>0.05);2The gene of MTNRIB such as rs1387153hasthe characteristic of variety performances.(1)The gene of CC, CT, TT in the rs1387153are allbased on the gene of C as an usual gene. The distribution of the genes in two teams fit into theregularity of Hardy-Weinberg.(2) The genic frequency of T2DM and compared groupCT,CC,AA separately are:50.80%vs49.50%,35.60%vs35.1%,13.60%vs15.40. Thedifference of1%have no statistics mean(P>0.05); The two group allelic gene’s frequency ofT separately are30%vs40.20%,61%vs59.8%. The difference also have no statistics mean(P>0.05);(3) The gene frequency and allelic gene frequency of T2DM and compared groupCT,CC,TT both have no marked difference(P>0.05);3、The polymorphism of MTNR1B geners1387153and clinical relation is:The T2DM patients TG with TT gene(P<0.05). TheT2DM patients SBP、DBP、HC、TC、、LDL、HDL with CT/TT gene have no marked differencecompared with the patients with PP gene (P>0.05). Conclusion:1、The T2DM of Ningxiaethnic Han may have relationship with the locus of MTNR1B gene rs1387153.
