Study Of The Association Of MTNR1B Rs1387153Gene And KLT14rs972283Gene Polymorphism With2Diabetes Mellitus In The Northern Area Chinese

Type2diabetes mellitus is one of the complex syndromes which caused by both ofhereditary factors and environmental factors, it has strong genetic susceptibility.Therefore we used PCR-RELP to detect the MTNR1B rs1387153and KLT14rs972283in1253patients with type2diabetes mellitus(T2DM)and1340control subjects withnormal glucose tolerance without family of T2DM in northern area Han nationalitypeople in order to investigate the association of the MTNR1B rs1387153and KLT14rs972283variant with T2DM.Objective: To investigate the association of the MTNR1B rs1387153and KLT14rs972283variant with T2DM.Method: Case group include1253patients with T2DM and control group include1340normal glucose tolerance subjects with fasting glucose less than5.6mmol/l andOGTT glucose less than7.8mmol/l in northern nationality were selected. The controlgroup was separed into age、 sex、 overweight subgroup (BMI≥24kg/m)andnon-overweight subgroup (BMI＜24kg/m).The DNA was isolated from peripheralblood cell according to standard procedures.Sense and anti-sense primes of the twoSNPs of the MTNR1B rs1387153and KLT14rs972283were designed according to thesequence of DNA.The PCR products were digested with restriction enzyne DdeI andXSPI for8hours,respectively. The genotypes were read after separation of the digestedproducts with8%SDS-PAGE.Different distribution in genotypes between T2DM andnon-diabetetic subjects was analyzed.We analyzed that the two SNPs composed withthe northern national people.Results: The four groups of MTNR1B rs1387153gene and KLT14rs972283genedid not showed statistically significant. Conclusion: There was no association between MTNR1B rs1387153gene、KLT14rs972283gene in the northern national people.
