Objective To investigate the association of visfatin gene single nucleotide polymorphism (-1535C/T) and its plasma levels with type 2 diabetes mellitus in Ningxia Hui and Han population.Method The study enrolled type 2 diabetes mellitus patients and healthy subjects of Hui and Han nationality population who have been lived in Ningxia province for three generations and haven't Han-Hui intermarriage history. Blood samples of type 2 diabetes mellitus patients (102 in Hui /108 in Han) and healthy subjects (101 in Hui /106 in Han) were drawn from endocrinology department of Affiliated Hospital of Ningxia Medical University and Ningxia People's Hospital. The plasma visfatin concentration was measured by enzyme-linked immunosorbent assay (ELISA).The visfatin gene(-1535C/T) polymorphisms genotypes were detected by PCR-RFLP. A set of examinations including blood pressure (BP),body mass index (BMI),waist hip ratio (WHR),fasting blood glucose(FBG),2-Hr postprandial glucose(2hPG)and lipids were carried out in all of subjects. The date was analyzed with statistics software of the SPSS 11.5, to calculate the distribution of allele and genotype frequencies of the visfatin gene polymorphisms in two groups of the Hui and Han nationality population of Ningxia, to test the association of visfatin gene single nucleotide polymorphism (-1535C/T) and plasma levels with type 2 diabetes mellitus in Ningxia Hui and Han population.Result (1)In Hui nationality population, the plasma visfatin concentration of 2 type diabetes mellitus group was between 8.34 and 48.39 ng/ml, the average value was(21.77±8.64)ng/ml; the plasma visfatin concentration of control group was between 3.59 and 25.59 ng/ml, the average value was (13.98±5.50)ng/ml. Plasma visfatin concentration in disease group was significantly higher than that of control group (P<0.05). BMI, WHR, SBP and the levels of FPG, 2hPG, HbA1C, TC, TG were higher in T2DM group than in normal control group (P<0.05). HDL-C was lower in T2DM group than in normal control group (P<0.05). The plasma visfatin concentration in type 2 diabetes was positively correlated with BMI, WHR and FPG.(2)In Han nationality population, the plasma visfatin concentration of type 2 diabetes mellitus group was between 6.18 and 45.27 ng/ml, the average value was (18.31±7.4)ng/ml; the plasma visfatin concentration of control group was between 3.00 and 25.63 ng/ml, the average value was (13.00±6.68)ng/ml. There was significant difference in the visfatin concentration between disease group and control group (P<0.05).WHRand the levels of FPG, 2hPG, HbA1C, TC, TG were higher in T2DM group than in normal control group (P<0.05). HDL-C was lower in T2DM group than in normal control group (P<0.05). The plasma visfatin concentration was positively correlated with WHR, FPG, 2hPG, HbA1C, and TG.(3) In Hui nationality population, the distribution of genotype of visfatin (-1535C/T) in case/control group was as follows: CC 19/20(18.6%/19.8%), CT 49/49(48%/48.5%), TT 34/32(33.4%/31.7); The distribution of the C and T allele in case/control group was as follows: C allele 87/89(42.6%/44.1%), T allele 117/113(57.4%/55.9%). The frequencies of the visfatin (-1535C/T) polymorphism were not different in diabetes and control groups(P>0.05).The frequencies of the visfatin (-1535C/T) polymorphism were not different in BMI≥25.0kg/m2 and BMI<25.0kg/m2(P>0.05). (4) In Han nationality population, the distribution of genotype of visfatin (-1535C/T) in case/control group was as follows: CC 20/25(18.5%/23.6%), CT 55/45(50.9%/42.4%), TT 33/36(30.6%/34); The distribution of the C and T allele in case/control group was as follows: C allele 95/95(44%/44.8%), T allele 121/117(56%/55.2%). The frequencies of the visfatin (-1535C/T) polymorphism were not different in diabetes and control groups(P>0.05).The frequencies of the visfatin (-1535C/T) polymorphism were not different in BMI≥25.0kg/m2 and BMI<25.0kg/m2(P>0.05).(5) In diabetes and control groups, the frequencies of the visfatin (-1535C/T) polymorphism were not different between Hui and Han nationality population(P>0.05).(6) In Hui disease group, the subjects who had TT genotype showed significantly lower serum HDL-C levels compared with those who had CC and CT genotype (P<0.05).In Han disease group, the genotype TT is higher than CC and CT in serum triglyceride(P<0.05).Conclusions (1) The levels of visfatin in type 2 diabetes group of Hui was higher than healthy control group. BMI, WHR, SBP and the levels of FPG, 2hPG, HbA1C, TC, TG were higher in T2DM group than in normal control group (P<0.05). HDL-C was lower in T2DM group than in normal control group (P<0.05). The plasma visfatin concentration in type 2 diabetes was positively correlated with BMI, WHR and FPG.(2) The levels of visfatin in type 2 diabetes group of Han is higher than healthy control group. WHR and the levels of FPG, 2hPG, HbA1C, TC, TG were higher in T2DM group than in normal control group (P<0.05). HDL-C was lower in T2DM group than in normal control group (P<0.05). The plasma visfatin concentration was positively correlated with WHR, FPG, 2hPG, HbA1C, and TG. (3) In diabetes and control groups, the frequencies of the visfatin (-1535C/T) polymorphism were not different between Hui and Han nationality population (P>0.05).The visfatin gene single nucleotide polymorphism (-1535C/T) were not associated with type 2 diabetes in Ningxia Hui and Han Population. The visfatin gene single nucleotide polymorphism (-1535C/T) were not associated with obesity in Ningxia Hui and Han Population.(4) The visfatin gene single nucleotide polymorphism (-1535C/T) were associated with abnormal serum lipids of type 2 diabetes in Ningxia Hui and Han Population.
