The Elymus dahuricus×Hordeum brevisubulatum F1 was used as experimental material. The regeneration system of Elymus dahuricus×Hordeum brevisubulatum F1 was established using young spike of Elymus dahuricus×Hordeum brevisubulatum F1. The callus and seedlings tillering stage were treated with colchicine to double chromosome. The main results were as follows:(1) The optimal medium for inducing callus was MS+3 mg/L 2,4-d+30 g/L sucrose+7 g/L AGAR powder.(2)The best differentiation medium for callus was MS+1 mg/L 6-B A+1 mg/L NAA+30 g/L sucrose+7 g/L AGAR powder.(3) The best rooting medium was 1/2 MS+0.5 mg/L IBA+0.5 mg/L NAA+30 g/L sucrose+7 g/L AGAR powder.(4) The best medium for callus subculture was MS+1 mg/L 6-BA+0.5mg/L2, 4-d+30 g/L sucrose+7 g/L AGAR powder.(5) The suitable colchicine concentrations for inducing chromosome doubling of tillering seedlings of Elymus dahuricus×Hordeum brevisubulatum F1 was 0.2%, the optimal treating time was 24 hours.(6) The optimal time treating callus of Elymus dahuricus×Hordeum brevisubulatum F1 with colchicine solution of 10 mg/L was 48 hours.
