Chunlan (Cymbidium goeringii) is a perennial herb. As one of the important species of terrestrial orchids, it not only has high ornamental value, but also has some medicinal value. The traditional plant breeding is inefficient. In order to protect the species resources of orchid, it has the very important meaning to study the technique of tissue culture of cymbidium. In this experiment, hybrid Chunlan as materials, we systematically studied the influence of various factors such as culture medium, hormone, carbon sources, additives on the germination and proliferation of cultivating materials. It providing a theoretical basis for industrialized breeding technology of Chinese orchid plant. The primary results were summarized as follows:(1) The best hormone ratio required for germination of hybrid Chunlan seeds was 0.5mg/LNAA+0.5mg/L6-BA. Adding 0.02% activated carbon and 0.1% tryptone could improve seed germination rate. Cultivating mode also had effect on seed germination rate, the germination rate of liquid culture was high than it in solid medium.(2) Basal medium of hybrid Chunlan rhizome proliferation cultured to 1/2MS as well. The best hormone ratio was 2.0mg/LNAA+0.1mg/L6-BA. Medium supplemented with sucrose was best to the rhizome proliferation. Natural compounds could significantly affect the culture of orchid material, the effect of tryptone promoting rhizomes proliferation was obviously better than that of coconut milk, apple Juice and banana juice. The comprehensive effect to rhizomes proliferation of adding 2.0mg/L tryptone was better than that of several other concentration levels. Activated carbon when the concentration of 1.0mg/L was favorable to the rhizome proliferation and activated carbon of high concentration could inhibit the growth of rhizome proliferation. The temperature in the 20~25 ℃ was appropriate to rhizomes proliferation. When the light intensity is 44001x the effect of rhizomes proliferation was better than that in 24001x, the optimum light intensity should be somewhere in between. The illumination time of rhizomes proliferation in the 10h was superior to 16h. The 60% of the relative humidity was suitable for the proliferation of orchid cultivation materials.(3) The optimum medium of inducing differentiation of hybrid Chunlan was 1/2MS+0.2mg/LNAA+2.0mg/L6-BA+0.2tryptone, and the differentiation process was accompanied by the proliferation and rooting. Seedling height above 10cm was more suitable than that of 1-3 cm in rooting culture.
