| Chenopodium quinoa is a typical halophyte,which has special biological characteristics and rich nutritional value.In recent years,people have a deeper understanding of quinoa.One of the major limitations to the large-scale cultivation of quinoa as a food crop is viral disease,its genetic composition of the population is heterozygous,and the heredity is unstable,these conditions greatly limited the spread of quinoa.It can be used to detoxify and preserve the excellent characters of the mother,provide technical support for the genetic transformation of quinoa and provide a reference for the growth and cultivation of adventitious buds after transgenic quinoa.In this paper,the seeds of"Shanyin 1"quinoa were used as materials,the best disinfection method was used to disinfect the seeds,and the aseptic seedlings were cultivated.In the process of the research on the micropropagation system of quinoa,the best conditions of explants selection,axillary bud and cluster bud induction,proliferation,strong seedlings,and root induction of test tube seedlings of quinoa were screened,and finally in vitro culture asexual propagation system of"Shanyin 1"quinoa was established.The results are as follows:1.The best disinfection method of quinoa seeds:the effect of 0.1%Hg Cl2disinfection time was studied.The results showed that the highest survival rate was observed when 0.1%Hg Cl2was used for 10 minutes,and the pollution rate was only5.26%.2.Selection of xenograft of quinoa:the callus and bud state induced by different explants were studied.The results showed that the best explant of callus induction is the embryo axis,the highest callus induction rate and the best growth state under the same treatment;the best explant for inducing axillary bud is cotyledons with 1 cm hypocotyl and apex bud;the best explant of inducing cluster bud is cotyledons with top bud.3.The best conditions for inducing,proliferating and rejuvenating of armpit buds of quinoa were studied.The effects of 6-BA,MS ions and p H value of different media on the induction,proliferation and rejuvenation of armpits of quinoa were studied.The results showed that the best medium for axillary bud induction was MS+6-BA3.0 mg/L,the axillary bud induction rate was 97.50%,the most suitable medium was2MS+6-BA 3.0 mg/L,the most suitable medium was 2MS+6-BA 0.1 mg/L,the number of plants in 1 cm accounted for 94.36%of the total plant number,and the optimal medium p H value was 6.0.4.The best conditions for inducing,proliferating and rejuvenating of the cluster buds of quinoa were studied.The effects of 6-BA,plant growth regulators of different types and concentrations on the induction,proliferation and rejuvenation of the cluster buds were studied.The results showed that the best medium for cluster bud induction was MS+6-BA 1.0 mg/L+NAA 0.1 mg/L,the rate of induction was 12.14%,the most suitable medium was 2MS+6-BA 0.5 mg/L,and the average number of cluster buds was 3.6 cluster buds per explant,the most suitable medium for the regeneration of cluster buds was 2MS+6-BA 0.3 mg/L,and the number of plants in the medium was 93.04%of the total plant number,and the p H value of the medium was 6.0.5.The best root induction conditions of quinoa plantlets were as follows:the effects of IBA at different concentrations on Rooting of the plantlets were studied.The results showed that the best medium for rooting of quinoa tube seedling was MS+IBA 1.0 mg/L,the rooting rate was 51.04%,and the number of main roots and roots>1 cm was the most.In this medium,the growth of adventitious root can be promoted effectively,the root system is thicker and the plant growth state is the best. |
