Periploca forrestii Schlt belongs to Asclepiadaceae family and distributes in the Yunnan-Guizhou Plateau, is widely used as herbal medicines by native people for the treatment of closed soft tissue injury, rheumatism, and rheumatoid diseases.Recently it had been developed to a variety of merchandise medicine, it was named as Periploca forrestii Schlt in Guizhou, while named Periploca sepeium in Yunman. In this study, We using Plant tissue culture techniques, we were able to put Periploca forrestii Schlt stems and terminal buds into MS medium with different concentrations of6-BA, NAA and2.4-D, and induced to complete plants, providing guidance for artificial propagation. At the same time.we took15Periploca sepium samples as the testing materials, which were sampled from different areas in Southwest China. Throughout the combination of40SRAP cited material, we yield total420amplification articles with analysis of NTSYS-the pc2.1software calculating the Jaccard’s genetic similarity coefficient and genetic distance. We also used UPGMA method to drawcluster map, in order to examice the genetic structure of the samples.The results are as following:①The best of Periploca forrestii Schlt terminal bud induction medium is MS+6-BA1.0mg/L+NAA0.3mg/L, in which bud induction rate reached86.29%; The best of Periploca forrestii Schlt stem segments induction medium is MS+6-BA2.0mg/L+NAA0.5mg/L, and the axillary bud induction rate can reach56.67%;②The optimum medium for the MS of proliferation is MS+6-BA2.0mg/L+NAA0.1mg/L, the multiplication coefficient can reach2.10; The best rooting medium was1/2MS+IBA0.5mg/L, rooting rate reached53.33%.③The polymorphism rate of Selected sepium materials was79.28%, genetic distance cariation were range from0.040-1.613, with an average of0.840, genetic similarity coefficient changed within0.391-0.973, the average was0.718.④SRAP markers are applicable to germplasm genetic diversity in Periploca forrestii Schlt. Test SRAP spectrum showed the Periploca forrestii Schlt in different regions of the respective cluster and revealing the origin and germplasm resources groups contact. No significant difference between the slopes with or without sunshine, however the influence of altitude is significant, with a degree of correlation in genetic distance and spatial distance.⑤Hei Gu-Teng and Dian Gang-Liu are both Chinese herbal medicines used Periploca forrestii Schlt, which were verified in the DNA level. Periploca calophylla(Wight.)Falc is far from Periploca forrestii Schlt in genetic, which are used as the alternatives among locals,.To ensure the safety and efficacy of the medicine, it is recommended to use Periploca forrestii Schlt as less as possible,with little or no alternatives.⑥We can build the SRAP pedigree of Periploca forrestii Schlt in each producing areas, which are used as a backup method to identify the different origin of Periploca forrestii Schlt.
