| The seven aspects of technology system about tissue culture and rapid propagation of Vietnam Mesona blumes were studied in this paper, including:(1)the test of explants sterilization;(2) the test of callus induction;(3) the test of callus differentiation;(4) the test of aseptic seedling bud proliferation;(5) the test of vitrification factors;(6) the test of aseptic seedling bud radication;(7) the test about transplanting the plantlets. The main results were as follows.1. During the explants sterilization test of Vietnam Mesona blumes, the test results showed that the effect of the combination0.1%HgCl2llmin+75%Ethanol55s on tender stems was optimum, the average pollution rate showed only3.17%.2. In the callus induction study of Vietnam Mesona blumes, the results showed that the effect of the grop MS+SUC25g/L+Agar6g/L+NAA0.1mg/L+2,4-D1.Omg/L to tender leaves piece displayed the best, callus induction rate reached100%and callus size peaked at0.72cm3.3. As for the callus differentiation test of Vietnam Mesona blumes, the outcome revealed that the callus, which was firstly induced by tender stems without bud in the induction medium group:MS+SUC25g/L+Agar6g/L+NAAO.lmg/L+6-BA1.Omg/L, and then transferd into the differentiation medium combination:MS+SUC25g/L+Agar6g/L+6-BA1.0mg/L+NAA0.3mg/L+KT0.5mg/L, after cultured for30days, the effect of callus differentiation come to the best and the bud differentiation rate was79.17%.4. Regarding aseptic seedling shoot proliferation test of Vietnam Mesona blumes, research has revealed that the seventh generation single bud, which came from stem upper position inoculated in the medium of MS with SUC20g/L, showed the best bud proliferation multiple. As for the different kinds of cytokinins, CPPU showed the better effect on bud multiplication than the others, at the same time, CPPU0.7mg/L showed the significant effect whether on before-effect to bud proliferation or on after-effect to bud proliferation. As to the different kinds of auxins, IAA0.05mg/L was the better choice than the others, with the bud proliferation multiple peaked at8.41.5. Regarding factors about aseptic seedling shoot vitrification test of Vietnam Mesona blumes, the results indicated it was the best that the effect of the combination of MS+SUC35g/L+Agar6g/L+6-BA0.1mg/L+NAA0.05mg/L+AC0.9g/L, the vitrification rate was only13.56%, with weak growth status.6. Regard to aseptic seeding radication test of Vietnam Mesona blumes, the stem upper position was the better for radication than stem upper position and the middle part. Comparison of the different auxins of NAA, IBA, ABT and IAA displayed that the radication result peaked in the group of1/2MS+SUC20g/L+Agar6g/L+IAA0.3mg/L, rooting rate was99.33%, average rooting number peaked at10.02and average root length was1.03cm. Comparison of the different concentrations of PP333showed that the rooting status was optimal in the combination of1/2MS+Agar6g/L+SUC20g/L+IAA0.3mg/L+NAA0.3mg/L+PP3330.6mg/L, rooting rate was100%, average rooting number peaked at23.65and average root length was0.33cm. Comparison of the different concentrations of B9revealed that in the combination of1/2MS+Agar6g/L+SUC20g/L+IAA0.3mg/L+NAA0.3mg/L+B90.4mg/L, rooting rate was100%, average rooting number reached27.63and average root length was1.24cm.7. In the transplanting plantlets test of Vietnam Mesona blumes, Perlite:peat(1:2) was the best transplanting matrix combination for transplanting plantlets, the survival ratio showed78.33%and the state of plant was the best.. |
