Armillaria mellea has high economic value that using their hyphae and rhizomorph for medicine. The growth of Polyporus and Gastrodia elata must rely on Armillaria mellea to provide nutrition.It is a unique geographical environment and climate in Qinba Mountain. And it is one of China’s largest main producing areas of Gastrodia elata. Now Armillaria mellea strains become serious degradation because of using year by year. To gain the excellent Armillaria become a major factor in the yield of Gastrodia elata. Collecting Screening, separating and purificating Armillaria from the main production areas of Gastrodia elata in Qinba mountainous. Selecting the good Armillaria strains to lay the foundation for the dustry of Gastrodia elata to technology and industrialization in Qinba mountains. The main results are as follows:First, 18 Armillaria strains were collected and obtained after separating and screening in the Qinba Mountains. Study on the Armillaria mellea by using PDA solid medium. The best growth characteristics of the Armillaria is ZY-3, ZY-5, ZY-9, followed by the ZY-15, ZY-17, others’ growth characteristics are not evident.Second, by extracting DNA, amplifying rRNA-ITS sequences of 18 Armillaria and comparing similarity with each other. The result shows that ZY-15 is significant species and different with others. By constructing phylogenetic tree for 18 Armillaria species, we get the identification and relationship category. 18 Armillaria strains were divided into four genera: Armillaria sinapina, Armillaria gallica, Armillaria mellea and Armillaria cepistipes. Preparating the crude enzyme solution, studies on esterase isozyme of Armillaria strains. By comparing the similarity coefficient among 18 strains, It shows that they have rich biological diversity.Third, by asexual reproduction and sexual reproduction test, the yield has significant differences between red and black Gastrodia elata. The results show that ZY-9, ZY-17 can significantly improve the yield of Gastrodia elata.Last, the dominant strains ZY-9, ZY-17 were tested by single factor and orthogonal experiment. Using biomass as a measurement index, the culture conditions of 2 strains of Armillaria mellea were optimized. Get the best carbon source is glucose, the best nitrogen source was peptone, the best inorganic salt is Na2HPO4.
