Abstract/Summary:
Gastrodia elata BI.is a precious Chinese medicinal material in China.In recent years,G.elata health care products have also been loved by people,and the market is in short supply.The two strains of G.elata directly affect the germination rate of G.elata seeds and the yield of G.elata.High temperature is an important factor affecting the two strains of G.elata.When the temperature is continuously higher than 30℃,the hyphae of the two strains of G.elata will lose vitality or even die.However,the high temperature phenomenon in the main producing areas of G.elata in China is serious in summer,and the high temperature weather above 30℃can last for more than ten days.At the same time,temperature is also closely related to the activity of extracellular enzymes secreted by fungi,which play an important role in their own growth and development.Therefore,it is very important to study the domestication of the two strains of G.elata resistant to high-temperature-resistant and the response of the extracellular enzyme activities of the two strains of G.elata to temperature,in order to promote the development and utilization of G.elata and the two strain industries.The main tests and results of the paper are as follows:(1)The two strains of G.elata preserved in the laboratory were domesticated by progressive high-temperature-resistant,and the two strains of G.elata that could grow at 34,26,28,30,30.5,31.5,31.5,32,32.5,33,33.5,34°C were gradually slowed down.The two strains of G.elata that could grow at 34°C were obtained,The high-temperature-resistant Mycena purpureofusca obtained by domestication were named:MDC-1,MDA-1,8C-1,SH-1,and the high-temperature-resistant Armillaria mellea were named:J-G,A-G,MH-G,and the test strains were determined for the follow-up test.(2)Analysis of the changes of extracellular enzymes in liquid culture of two strains of G.elata.It is speculated that the M.purpureofusca mainly use cellulose and lignin analogs in the early stage;cellulose and hemicellulosic substances are mainly used in the middle stage;cellulose,pectin and starch substances are mainly used in the later stage.The A.mellea mainly used lignin analogs in the early stage;pectin,starch and lignin substances are mainly used in the middle stage;hemicellulose and lignin substances are mainly used in the later stage,cellulose and pectin substances are mainly used in the final stage.(3)Set the culture temperature of 25°C and 30°C each for 8 h cycle,determine the extracellular enzyme activity and G.elata seed germination rate,the domesticated high-temperature-resistant M.purpureofusca 5 extracellular enzyme activities and G.elata seed germination rate are significantly higher than the corresponding starting strains(P<0.01),hydrolyzed plant cell wall pectinase,xylanase and their active size are the key extracellular enzymes affecting the germination of G.elata seeds,the correlation coefficients are 0.883,0.720,high-temperature-resistant M.purpureofusca SH-1、MDA-1 G.elata seed germination rate is the highest,germination rate are 71.76,68.82%.(4)The cultivation verification test of G.elata was carried out,and the high-temperature-resistant M.purpureofusca after domestication promoted the germination of G.elata seeds under high-temperature conditions in summer,and the high-temperature-resistant A.mellea strains MH-G and J-G after domestication promoted the growth of Gastrodia elata under high temperature conditions in summer,and the combination of high-temperature-resistant A.mellea MH-G and high-temperature-resistant M.purpureofusca SH-1 was the best,with a fresh weight of 128.80 g/bag and a dry weight of15.20 g/bag.The results of different cultivation methods of G.elata showed that the average germination rates of M.purpureofusca SH、MDA and MDC in bag cultivation were 1.32,6.09,3.67%higher than those in the same combination field,respectively;The average fresh weight of A.mellea A-10,A-G,J-234,J-G,MH-425 and MH-G planted in bags was higher than that of a field planted,respectively:2.23,0.27,0.33,0.56,0.31,3.50 kg/m~2.
